Identification and characterization of a polyurethanase with lipase activity from Serratia liquefaciens isolated from cold raw cow's milk

“…According to the amino acid sequence alignment of AprF in the BL21(DE3) genome (KEGG entry number T02096, GenBank accession number AM946981) by tblastn of BLAST at the NCBI, the ABC exporter with TolC (B21_02857) was deduced to assist the extracellular secretion of PueA and PueB. Similarly, PueA and PueB from P. chlororaphis have also been reported to be directly secreted into the extracellular environment in E. coli XL1-Blue MRF' (Stern and Howard 2000;Howard et al 2001), and a single glycine-rich repeat of PueA (aa 349-402) can mediate the secretion of a GST fusion protein in XL1-Blue MRF' independent of an additional ABC exporter (Langlois and Howard 2002). Moreover, subfamily I.3 lipases LipPS1 from P. synxantha PS1 (Cai et al 2016) and LipI.3_KE38 from P. fuorescence KE38 (Karakas ß and Arslano glu 2020) are also released into the extracellular environment in the form of direct secretion in BL21(DE3).…”
