Human natural killer cells for targeting delivery of gold nanostars and bimodal imaging directed photothermal/photodynamic therapy and immunotherapy

Abstract: Objective: To construct a novel nanoplatform GNS@CaCO3/Ce6-NK by loading the CaCO3-coated gold nanostars (GNSs) with Chlorin e6 molecules (Ce6) into human peripheral blood mononuclear cells (PBMCs)-derived NK cells for tumor targeted therapy. Methods: GNS@CaCO3/Ce6 nanoparticles were prepared and characterized by TEM and UV-vis. The cell surface markers and cytokines secretion of NK cells before and after loading the GNS@CaCO3/Ce6 nanoparticles were detected by Flow Cytometry (FCM) and ELISA. Effects of the GN… Show more

“…Smokers have increased plasma concentrations of various toxic heavy metals, which are responsible for the genotoxic effects of cigarettes on human DNAs by inducing double‐strand breakage and inhibiting DNA repair 39 . The protective effect of GNPs can be explained by the interaction between cellular constituents and GNPs; when reduced by citrate, GNPs actively interact with leukocytes, erythrocytes, and platelets to increase adsorption 40,41 . This beneficial interaction between GNPs and defense cells, called hemocompatibility, forms a corona enriched with proteins involved in humoral immunity and increases the ability of defense cells to fight pathological agents and repair damaged cells 40 .…”

“…Based on the traditional seed method, further integration and chemical modification of AuNSs with other components leads to the creation of various kinds of AuNSbased nanohybrids, such as metal compound/AuNSs, 28 SiO 2 /AuNSs, 30 dendrimer/AuNSs, 20 and MOF/AuNSs nanohybrids. 22 For instance, Montoto et al 30 reported the preparation of mesoporous silica-coated AuNSs.…”

“…NK cells expansion in genetically engineered K562 cells was performed from umbilical cord blood mononuclear cells (UB-MNCs). Irradiated K562 feeder cells were received from Hangzhou Zhongying Bio-Medical Technology (Zhejiang, China), which coexpress membrane-bound IL-21 and CD137L (4-1BBL) 23 . On day 0, UB-MNCs and 2 × 10 7 genetically engineered K562 cells (K562-mb215-41BBL, Zhongying Bio-Medical Technology) were resuspended in 40 ml NK cell medium [containing 90% RPMI-1640 HyClone, Logan, UT, USA; 10% fetal bovine serum, Gibco, Carlsbad, CA, USA; and 100 µg/ml IL-2, R&D, Minneapolis, MN, USA].…”
