ABSTRACT:The aim of this study was to evaluate the effects of immunising farm animals with the Histophilus somni recombinant heat shock protein 60kDa (H. somni rHsp60) in field conditions. Fifty piglets, 10 calves and 30 hens were immunised twice, and the same number of each species was used as the control. The humoral immune response was evaluated using ELISA in piglets (IgG, IgA and IgM) and calves (IgG 1 , IgG 2 and IgM) sera and in hen egg yolks (IgY). Cell-mediated immune responses were evaluated using the skin test. Concentrations of serum haptoglobin in calves and piglets and plasma fibrinogen in calves, daily weight gain in piglets, as well as the inner body temperature and clinical signs in calves were measured to evaluate the clinical effects of vaccination. In animals immunised twice with H. somni rHsp60, a statistically significant increase in IgY antibodies in egg yolk as well as serum IgG 1 and IgG 2 antibodies in calves (P < 0.05) was found. In piglets, the antibody reaction against H. somni rHsp60 was higher in the experimental than in the control group, but the difference was significant only for the IgG class (P < 0.05). A moderate cell-mediated immune response to H. somni rHsp60 measured using the skin test was observed in piglets after 24 h (P < 0.05), but not in calves and hens. The daily weight gain was significantly higher in the experimental than in the control piglets (P < 0.05). The fibrinogen and haptoglobin levels in calves, as well as the inner body temperature, indicated a reduced risk of pathology in the experimental group of calves. The preliminary results confirmed the immunogenicity of H. somni rHsp60. A beneficial effect on piglet weight gain was observed. The obtained results warrant further studies of the protective effects of H. somni rHsp60 as an ingredient of subunit vaccines in farm animals.

Hsp60 of Salmonella Enteritidis appears to be involved in pathogenesis of infectious processes and host immune responses. Because of the similarities between microbial and human Hsps, a humoral response against microbial Hsps may be destructive for the host due to antigen mimicry leading to an autoimmune response. We have performed a preliminary study to investigate whether the homology between Hsp60 of Salmonella Enteritidis and human Hsp60 really exists. ELISA tests were done with success to show that polyclonal antibodies developed against Hsp60 of Salmonella Enteritidis recognize and cross-react with human Hsp60 and one of its seven synthetic peptides used in the study. It was revealed that the antigenic epitopes, which both the proteins have in common, are located between 409 and 424 amino acid residues of human Hsp60 molecule, and are determined by the amino acid sequence of human Hsp60 (409-424) synthetic peptide, the same sequence which was found by other authors to be an immunodominant epitope in patients with acute coronary syndromes. The results show the presence of an immunological and sequence similarity between bacterial Hsp60 and its human counterpart, and suggest that human Hsp60 could be a possible target of immune response triggered by Hsp60 of Salmonella Enteritidis. Salmonella Enteritidis Hsp60 might be potentially involved in autoimmune mechanisms operating in humans. This conclusion must be considered preliminary and hypothesis-generating than hypothesis-proving.

The present study aims at developing an indirect ELISA to quantify yolk antibodies specific to all surface proteins of the invasive Salmonella Enteritidis (SE), which acquired the 1.8, 14.1, and ~ 50 Kb plasmids. An ELISA checkerboard was used in four different experiments to account for the different parameters included in the preliminary ELISA procedure, and consequently to maximize the difference in Optical Density (OD) values between control positive and negative yolk samples. The first experiment aimed at studying the impact of 5% Bovine Serum Albumin (BSA) dissolved in distilled water as a blocking reagent on a 28 µg/well SE antigen-coated plate, while applying the positive and negative control yolk samples to different concentrations of Phosphate-Buffered Saline (PBS).Conjugate application was maintained constant at a dilution of 1:500 in PBS. The second experiment was similar to the first one, but the positive and negative control yolk samples were diluted in PBS-Tween 20, and the conjugate dilution was changed to 1:1500 in PBS-Tween 20. In the third experiment, the conjugate was diluted at 1:1500 in 5% BSA/PBS-Tween 20 diluent or PBS-Tween 20 diluent with no 5% BSA. The objective of the fourth experiment was to study the impact of four different concentrations of SE-coated antigen levels (28µg/well, 56µg/well, 84µg/well, and 112µg/well), while fixing the blocking step with 5% BSA in distilled water, and the conjugate dilution set at 1:1000 in 5% BSA/PBSTween 20, and fixing the control yolk samples dilution at 1% in PBS-Tween 20. This last experimental procedure allowed the highest difference in mean absorbance OD values of the positive control minus the negative control samples, which was equivalent to 0.381. In addition, the final protocol for this ELISA was applied on individual egg yolk samples of two groups of chicken layers: one challenged in the esophagus at 11 days with 5.4 x 10(10) CFU/ml/bird of SE, and the second group was not challenged. The mean OD values of the egg yolk of antibodies specific against SE of the two groups were significantly different (0.8578 versus 0.5250; p<0.05), which indicates the possible application of the developed ELISA for screening SE infection by examining egg yolks produced by commercial layers
