AIM: To evaluate the immunomodulatory effects of ethanolic extract of herb pugun tanoh on TCD4 and TCD8 cells in Doxorubicin-induced rats. METHODS: Fifteen male Sprague Dawley rats were divided into five groups consisting of six rats each as follows: Group 1, DOX-treated rats (4.67 mg/kg BW body weight on day 1 and 4) and were administered normal saline 0.9% orally once daily for 7 consecutive days, Group 2, receiving Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) of dose 150 mg/kg BW orally, Group 3, receiving dose Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) 300 mg/kg BW orally. The rats of group 2-3 were intramuscularly administered with doxorubicin at a dose of 4.67 mg/kg BW at the days 1-4 to suppress immune functions. RESULTS: Treatment of 300 mg/kg BW of Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) succeeded in reducing side effect doxorubicin based on increasing the TCD4+ and TCD8+ blood level. CONCLUSION: Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) could increase the level of TCD4+ and TCD8+ in rats which induced by doxorubicin.

BACKGROUND: Pugun tano extract had been studied for its effect as hepatoprotector. However, the usage of the plant in the form of extract has a limitation, especially if the extract is consumed by the people due to the unpleasant taste and odour. Then, the extract needs to be transformed into a particular dosage form, such as a capsule. But before the capsule can be produced, a preformulation study of pugun tano extract into a granule mass in capsule need to be evaluated. AIM: The study aimed to formulate the ethanolic extract of pugun tano (Curanga fel-terrae (Lour.) Merr) as granule mass in the capsule dosage form. METHODS: The pugun tano ethanolic extract was formulated in several steps included preparation of dry extract using coating method with polyvinylpyrrolidone (PVP) and granule mass production. The excipients used for the granule mass were lactose granules (made with tapioca starch using wet granulation), corn starch (made with 3 concentrations of 5% (F1), 7.5% (F2) and 10% (F3)), talcum, magnesium stearate, methylparaben, and propylparaben. The granule mass was evaluated for the bulk density, tapped density, inter-particle porosity, Carr’s index, Hausner ratio, angle of repose, and flowability. RESULTS: The results showed that all of the formulae passed the requirement of the preformulation test. The bulk density of the granule mass was 0.79 – 0.86 g/ml; the tapped density was 0.88 – 0.90 g/ml; the inter-particle porosity was 0.03 – 0.14; the Carr’s index was 2.71 – 11.94%; the Hausner ratio was 1.09-1.12; the angle of repose was 26.10 – 28.90°; and the flowing time was 5.97 – 6.63 seconds. All of the formulae showed good flowability and free-flowing properties. CONCLUSION: It is concluded that the obtained formula

The objective of this study is to determine phytochemical constituent in the ethanol extract of Picria fel-terrae leaves as well as to examine the teratogenic effect toward the fetus. There are five groups consists of pregnant rats. Group 1 was the healthy control; groups 2, 3, 4 received extract at the dose of 125, 500 and 1000 mg/kg respectively, group 5 was positive control group (received gabapentin 50 mg/kg). The treatment was performed during the organogenesis period. On the 19 th day, maternal laparotomy was carried out, and the fetus was removed. Observations were including reproduction and uterus performance as well as external malformation.Phytochemical screening revealed that the extract consists of flavonoids, tannins, glycosides, saponins and steroids/triterpenoids. Administration extract at the dose of 125, 500, 1000 mg/kg revealed significant effect to the weight loss, fetal body length, number of dead fetuses, resorption and haemorrhages compared to healthy control (p<0.05). The uterus abnormality was found in the group of extract dose of 500 mg/kg as well as 1000 mg/kg. All groups undergo external malformations. The research concluded that all doses of the ethanol extract of P. fel-terrae leaves had the teratogenic effect on the fetus.
