HEMOPOIETIC STEM CELL PROLIFERATION AND MIGRATION FOLLOWING BORDETELLA PERTUSSIS VACCINE

Abstract: The ability of a single injection of killed, intact bacteria to effect an increase in the proliferative rate of hemopoietic stem cells was studied. The total numbers of colony forming units in bone marrow, spleen and peripheral blood as well as the proportion of CFU in cycle was assessed. Splenic CFU were observed to rise exponentially due initially to in situ proliferation and later to proliferation in bone marrow with migration via the blood to the spleen. The results are discussed in the light of current co… Show more

“…If this is so, the C. parvurn-induced rise in the (Table V). Similar results have been reported by Monette et al (1972) in B. pertussis-treated mice. This finding indicates that the CFUs accumulating in the spleen should be of exogenous origin, derived probably from bone marrow.…”

“…of 20X109 heat killed B. pertussis organisms. In normal mice, this treat ment effects a rapid fall of marrow erythroid precursors and a sharp in crease in the number of circulating C F U [7], In the present study, a 6.5-fold increase in blood C F U concentration in + / + mice was observed, while in Sl/Sld mice there was only a 2.4-fold increase (table I). The ob served increase in + / + mice was distributed among the various colony types.…”

“…Thus, in re sponse to the increased demand for certain blood cells, hemopoietic stem cells are mobilized via the bloodstream from bone marrow to various extra medullar hemopoietic organs where they undergo growth/differentiation. Mobilization of CFU-S into the circulation in response to the increased demand for neutrophils was already noted in mice treated with pertussis vaccine [15] or with bacterial endotoxin [16].…”
