Intensive shrimp culture has encountered many problems, such as declining water quality through disease caused by pathogenic microbes, which affected mortality. This study aimed to determine any potential probiotic from Bacillus sp. collected from mangrove in East Lampung, which could be used to improve the cultured shrimps' proteolytic and probiotic activity. This is a descriptive research with sampling and data collection of bacteria from many samples of mangrove. Result shows 128 isolates Bacillus from which then it has arrived at five potential probiotic Bacillus sp. The study five Bacillus sp. has been isolated with potential properties for probiotic (KPP212, IP121, UJ131, UJ132, SB141). Each isolate has characteristics with proteolytic property, growth in a wide range of pH 4–10 and osmotic stress (0–6% NaCl), non-pathogenic, ability for glucose fermentation, non-motile, and has negative catalase activity. The five potential Bacillus sp. can be used as probiotics for shrimp farming.

Abstract. Arifiyanto A, Afriani H, Putri MH, Damayanti B, Riyanto CL. 2021. The biological prospective of red-pigmented bacteria cultured from contaminated agar media. Biodiversitas 22: 1152-1159. Contaminated agar media was often depleted due to the growth of microbes, which is undesirable for culture. However, the contaminating microbes usually have a distinctive morphology. This research aimed to identify the potential of red-pigmented bacteria originated from the contamination of Drosophila melanogaster larvae. Fruit fly larvae that grow on contaminated tryptic soy agar were accompanied by the appearance of the red-pigmented bacteria colony. The bacterial colonies were purified by the re-streaking method on tryptic soybean agar. This strain was characterized morphologically, biochemically, and molecularly. Results showed that the MBC1 strain was identified as Serratia marcescens. The various metal susceptibility tests at 25 ppm did not affect the growth of the MBC1 strain. Meanwhile, it was also able to inhibit the growth of certain pathogens such as Aspergillus niger, Candida sp., Fusarium sp., and Rigidoporous sp. Strain MBC1 was able to produce antioxidant compounds. The lipolytic and amylolytic activity can be developed for bioremediate agriculture waste and biosurfactant production.

AB8 isolated from Lapindo Mud Volcano Area, Sidoarjo, Indonesia. Biodiversitas 22: 2817-2823. Streptomyces is a potential bacterial genus that has been investigated extensively as a source of natural microbial compounds. Its potential metabolites have been widely developed for pharmaceutical, pathogen control, and other applications in agriculture. This study aimed to determine the ability of the Streptomyces sp AB8 crude extract in inhibiting Plasmodium and pathogenic microbes. Streptomyces was cultured on Gause synthetic media for 10 days. The fermented broth culture media has dissolved in a 1:1 mixture of ethyl acetate and methanol. Biochemical characterization of this isolate has carried out using the standard methods. In-vitro antimalarial activity assay was performed using a chloroquine-sensitive Plasmodium falciparum strain 3D7. Fresh type O-positive human erythrocytes were suspended at 4 percent hematocrit in a complete medium to maintain culture. The inhibitory concentration (IC50) was determined using probit analysis. The results showed the extract of Streptomyces sp. AB8 contains phenolic and alkaloids. Streptomyces sp. AB8 extract can inhibit Dickeya zeae N-Unila 5, Dickeya zeae N-Unila 10, Aspergillus sp IK3, and Escherichia coli growth. The results also showed that the ICs0 value of extract against P. falciparum 3D7 was 17.56 ug/mL. Further research was needed to determine the types of purified bioactive compounds and their bioactivity.
