Hallmarks of Cancer: New Dimensions

Abstract: The hallmarks of cancer conceptualization is a heuristic tool for distilling the vast complexity of cancer phenotypes and genotypes into a provisional set of underlying principles. As knowledge of cancer mechanisms has progressed, other facets of the disease have emerged as potential refinements. Herein, the prospect is raised that phenotypic plasticity and disrupted differentiation is a discrete hallmark capability, and that nonmutational epigenetic reprogramming and polymorphic microbiomes both constitute di… Show more

“…The histopathologic tumor stage was pT2c for 47.9% of the patients, while 26.1% had pT3, and the ISUP grade group was 1 or 2 for 73.8% of the patients. The median CAPRA-S score was 3 (39.4% Capra-S low (0-2), 46.5% Capra-S intermediate (3)(4)(5), and 14.1% Capra-S high (6)(7)(8)(9)(10)(11)(12). Positive surgical margins were found in 30.5% of the cases.…”

“…The issue was revisited in 2011 [ 14 ], when four new hallmarks (not shown in the figure) were added to the list. In 2022, the list of hallmarks has increased up to 14 [ 19 ].…”

“…Cancers occur in an extraordinary range of types and subtypes, making each cancer individually unique [1]. Genetic and phenotypic differences are detected between tumors from different tissues and cell types, as well as between individuals with the same tumor type, a phenomenon known as inter-tumor heterogeneity [2].…”
