Halilectin 1 (H‐1) and Halilectin 2 (H‐2): two new lectins isolated from the marine sponge Haliclona caerulea

Abstract: Two new lectins named Halilectin 1 (H-1) and Halilectin 2 (H-2) were isolated from the marine sponge Haliclona caerulea using a combination of affinity chromatography on stroma fixed onto Sephadex G-25 and cation and anion exchange chromatography. H-1 is a monomeric protein with a molecular mass of 40 kDa estimated using sodium dodecyl sulfate polyacrylamide gel electrophoresis and 15 kDa estimated using a TSK gel. Conversely, H-2 is a homodimeric protein with 15 kDa monomers linked via weak interactions. H-1 … Show more

“…Similar to lectins isolated from Haliclona caerulea (Carneiro et al 2013), Axinella corrugata (Dresch et al 2011) and Aplysia kurodai (Kawsar et al 2010), HGL showed a high level of cytotoxicity against Artemia nauplii. However, hemolysis caused by lectin is rare, and few lectins have shown hemolytic activity (Hatakeyama et al 1994, Sudhakar andVincent 2014).…”

“…Presence of a non-toxic lectin from Sebastiania jacobinensis bark to Artemia nauplii is also reported (Vaz et al, 2010). Even two lectins (Halilectin 1 and Halilectin 2) from the same source, a marine sponge, had been purified with quite different toxicities (6.4 and 142.1 μg/ml) (Carneiro et al, 2013). Comparing different LC 50 values of these lectins, it became evident that STL-S and STL-D exhibited an intermediary toxic effect against Artemia nauplii.…”
