The dermatophytes are among the most frequently observed organisms in biomedicine, yet there has never been stability in the taxonomy, identification and naming of the approximately 25 pathogenic species involved. Since the identification of these species is often epidemiologically and ethically important, the difficulties in dermatophyte identification are a fruitful topic for modern molecular biological investigation, done in tandem with renewed investigation of phenotypic characters. Molecular phylogenetic analyses such as multilocus sequence typing have had to be tailored to accommodate differing the mechanisms of speciation that have produced the dermatophytes that are commonly seen today. Even so, some biotypes that were unambiguously considered species in the past, based on profound differences in morphology and pattern of infection, appear consistently not to be distinct species in modern molecular analyses. Most notable among these are the cosmopolitan bane of nails and feet, Trichophyton rubrum, and the endemic African agent of childhood tinea capitis, Trichophyton soudanense, which are effectively inseparable in all analyses. The molecular data require some reinterpretation of results seen in conventional phenotypic tests, but in most cases, phylogenetic insight is readily integrated with current laboratory testing procedures.

We have shown that dermatophyte species can be easily identified on the basis of a DNA sequence encoding a part of the large-subunit (LSU) rRNA (28S rRNA) by using the MicroSeq D2 LSU rRNA Fungal Sequencing Kit. Two taxa causing distinct dermatophytoses were clearly distinguished among isolates of the Trichophyton mentagrophytes species complex.Dermatophytes are the main cause of superficial mycoses (9,15,16). These fungi have the capacity to invade keratinized tissue of humans or animals to produce infections that are generally restricted to the corneocytes of the skin, hair, and nails. Among the approximately 10 species isolated in Europe, Trichophyton rubrum and T. mentagrophytes are the most commonly observed, with frequencies varying from 27 to 74% and from 17 to 41%, respectively (13).Dermatophytes are usually identified on the basis of macroscopic appearance, together with microscopic examination of cultures. Important characteristics are the rate of growth, the shape and texture of the culture on solid media, color, diffusion of pigments into agar, and sporulation. However, identification of dermatophytes often remains difficult or uncertain because there are variations from one isolate to another. Recent advances in molecular biology and progress in technology have allowed the development of new techniques for species determination and strain typing in microbiology. The molecular approach used to identify fungi is often based on sequence analysis of the ribosomal DNA (rDNA) and in particular on the internal transcribed spacer (ITS). The polymorphism of the ITS1 and ITS2 regions flanking the DNA sequence encoding the 5.8S rRNA was previously shown to be suitable for the identification of clinically important yeasts (1), Aspergillus sp. (8),and dermatophyte species (3-5). In contrast, the gene coding for the small-subunit rRNA (18S rRNA) did not discriminate sufficiently between dermatophyte species (7).The MicroSeq D2 large-subunit (LSU) rRNA Fungal Sequencing Kit (Applied Biosystems, Rotkreuz, Switzerland) was recently developed to identify fungal species after amplification of a partial sequence of the DNA encoding the LSU rRNA (28S rDNA). The sequence of a given fungus can then be compared for identification with the rDNA sequences of the MicroSeq D2 Fungal database, which includes more than 500 validated sequences from different fungal species but not from dermatophytes. In the present study, we tested the MicroSeq D2 LSU rRNA Fungal Sequencing Kit by using it to identify dermatophyte species from patients referred to the mycological laboratory of the Department of Dermatology at the University Hospital in Lausanne, Switzerland (Table 1). The presence of dermatophytes in animals with skin lesions (ringworm), as well as in animals with apparently healthy skin (healthy carriers) was studied. Skin scales and infected hair from 125 cats with skin lesions were examined microscopically by using 20% KOH and the methylene blue stain. Specimens were also inoculated in Sabouraud-chloramphenicol-cycloheximide agar. The hairbrush technique was performed in 200 cats with apparently healthy skin (120 of the urban and 80 of the rural area). Dermatophytes were isolated from 92 cats with skin lesions. M. canis was isolated from the 97,8% and T. mentagrophytes from 2,2% of the infected cats. Most of the infected animals (63) were up to 24 months old but the proportion between infected males and females cats was approximately equal. From the 120 cats of the urban area and the 80 cats of the rural area ( both groups included as apparently healthy cats) 40 (33,3%) and 15(18,7%) were found carrying dermatophytes, respectively. M. canis was exclusively isolated from the cats of the urban area ίη 70% while T. mentagrophytes in 15% and M. gypseum in 12,5%. The most frequent dermatophyte isolated from the apparently healthy cats of the rural area was M. gypseum (53,3%), while T. mentagrophytes (33,3%) and T. terrestre were the second and the third isolated. It is mentioned that infected cats and especially apparently healthy cats but carriers of M. canis, interfere with public health because of the possibility of transmission to human and given that numerous cases of children and of the animal owners have been affected.
