Gold Nanorod-based Photo-PCR System for One-Step, Rapid Detection of Bacteria

Abstract: The polymerase chain reaction (PCR) has been an essential tool for diagnosis of infectious diseases, but conventional PCR still has some limitations with respect to applications to point-of-care (POC) diagnostic systems that require rapid detection and miniaturization. Here we report a light-based PCR method, termed as photo-PCR, which enables rapid detection of bacteria in a single step. In the photo-PCR system, poly(enthylene glycol)-modified gold nanorods (PEG-GNRs), used as a heat generator, are added into… Show more

“…NPs have also been applied with tremendous success in biodetection systems, namely as sensors and diagnostics platforms with increased sensitivity and selectivity. Due to the decrease in size of the transduction mechanisms provided by NPs, most of these platforms have found applications at point-of-need and/or point-of-care (Costa et al, 2014 ; Veigas et al, 2014 ; Weng et al, 2015 ; Kim J. et al, 2017 ; Wang et al, 2017b ; Galvan and Yu, 2018 ; Yang et al, 2018 ). In some cases, diagnostics/sensing and therapeutic properties have been combined onto single NPs, providing for innovative tools – Nanotheranostics.…”

“…Plasmonic light-to-heat energy conversion contributed to a breakthrough ultrashort polymerase chain reaction cycle time of 30 cycles in 54 seconds [218]. This has induced more modifications with various plasmon generation methods, such as those involving the use of nanorods [219], Pt thin films [220], and Au thin films [221]. Plasmonic light-to-heat energy conversion is beneficial for rapid and precise heating in polymerase chain reaction.…”

“…It exploits the light-to-heat conversion mediated by plasmonic substrates [26][27][28][29] . This concept has been adopted for PCR thermocycling, in the form of a planar plasmonic substrate in a fluidic device 20,30,31 or metallic nanoparticles in suspension 22,32,33 . Both methods have shown fast nucleic acid ampli-fication, but challenges remain: nucleic acids after amplification are difficult to detect because plasmonic substrates would interfere with fluorescence reporters 34,35 .…”
