Glycosylation controls cooperative PECAM-VEGFR2-β3 integrin functions at the endothelial surface for tumor angiogenesis

Abstract: Most of the angiogenesis inhibitors clinically used in cancer treatment target the vascular endothelial growth factor (VEGF)/VEGF receptor (VEGFR) pathway. However, the current strategies for treating angiogenesis have limited efficacy. The issue of how to treat angiogenesis and endothelial dysfunction in cancer remains a matter of substantial debate. Here we demonstrate a glycosylation-dependent regulatory mechanism for tumor angiogenesis. St6gal1 mice, lacking the α2,6-sialylation enzyme, were shown to exhib… Show more

“…α2,6-sialylation mediates the homophilic interaction of the platelet endothelial cell adhesion molecule (PECAM). Sialylated PECAM interacted with two other sialylated receptors, VEGFR2 and integrin β3, while the inhibition of sialylation in ST6GAL1 −/− mice prevented PECAM-VEGFR2 interaction on the endothelial surface, inducing endothelial cell apoptosis and inhibiting angiogenesis [83].…”

“…N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (Neu5Gc) are the two major sialic acids in mammals [3]. Because of their negative electric charge, sialic acids play crucial roles in a variety of cellular functions, e.g., cell-cell interaction, determining conformation of glycoproteins on cell membranes, and masking antigenic determinants on receptor molecules [4][5][6][7][8][9][10][11].…”

“…When human WM793 primary melanoma cells gain metastatic competence, they show increased levels of sialylated αVβ3 integrins [77]. Cooperation of PECAM-VEGFR2-integrin β3 is significantly reduced when integrin β3 lacks 1,6-sialic acid [78].…”
