Gingival Crevicular Fluid Levels of Sclerostin, Osteoprotegerin, and Receptor Activator of Nuclear Factor‐κB Ligand in Periodontitis

Abstract: The GCF sclerostin level may be more reliable than the RANKL/OPG ratio as a diagnostic and prognostic marker of periodontal disease and treatment outcome. Regulation of sclerostin levels may aid the development of new therapeutic strategies for the treatment of periodontal disease.

“…The mRNA and protein levels of SOST were statistically significantly up‐regulated in the gingival tissue and serum samples of patients with chronic periodontitis when compared to periodontally healthy patients. Chronic periodontitis patients have also shown increased SOST levels in GCF in a prospective cohort study from Turkey, while non‐surgical periodontal therapy resulted in decreased levels which was related to clinical improvement of the periodontal status . Although the study of Balli et al also used GCF samples, it did not completely agree with our findings.…”

“…GCF concentration of OPG was lower in chronic periodontitis, but increased after periodontal phase I therapy, although not significantly (31). The RANKL/OPG ratio was also reported unchanged at 2, 3, and 4 months after periodontal therapy (33,34). In accordance with these studies, the RANKL value was not altered in any of the treatment periods in the healthy periodontitis group (group B).…”

“…Three hundred and fifty microliters of phosphate‐buffered saline (pH 7.4) was added to tubes with the sample strips. Concentrations of VEGF‐A,** TNF‐α, †† and endocan ‡‡ in GCF and serum were determined using a sandwich enzyme‐linked immunosorbent assay method according to the protocol provided by the manufacturer, as described previously 35 . Concentrations in GCF and serum samples were determined from standard curves.…”
