Genistein modulates gene activity in psoriatic patients

Abstract: Despite the impressive advancements in the treatment of psoriasis over the past two decades, there is still a need for further improvement. As previously shown in the literature, genistein (5,7-dihydroxy-3-(4-hydroxyphenyl)chromen-4-one), naturally occurring plant compound displays multidirectional action, also in relation to alleviating psoriasis symptoms. In this work we focused our attention on genistein impact on expression of genes when treating moderate-to-severe psoriasis patients. Testing the effects o… Show more

“…This outcome resulted in the inability to conduct any correlation analysis of inflammatory biomarkers with the clinical parameters of patients participating in this trial. Of course, clinical efficacy was rather poor in our test, which may be related to the fact that high doses of the agent do not result in better effects than low doses due to the limited absorption of isoflavone preparations 16 . In addition, frequent and modest intakes of genistein over the day rather than as a single high dose per day could be more efficient 34 .…”

“…(5,7-dihydroxy-3-(4-hydroxyphenyl) chromen-4-one), natural isoflavone, impaired the activity of certain genes which are overexpressed in psoriasis. It stimulated the expression of other genes that are repressed in dermatosis in both skin specimens and peripheral blood cells of psoriatic patients 4 .…”

“…Various plants from traditional Ayurveda, Siddha, Uighur, Thai, Korean, Chinese medicine are used for the management of various skin ailments, psoriasis, vitiligo, dermatitis, and leucoderma. A large population uses complementary medicines for two reasons, namely, the limitations of synthetic drugs and the ability of the natural compounds to act on multiple targets 4 . The plants can pave the way for new therapeutic approaches to psoriasis treatment since they have significant preventive activity against psoriasis 5 .…”

“…It is thus supposed that genistein may attenuate ROS-mediated NF-κB activation and NF-κB-dependent inflammatory cytokine production in psoriatic lesional keratinocytes. Oral administration of genistein in psoriasis patients for 8 weeks impaired the transcription of genes overexpressed in psoriasis, CXCL10 , IL-6 , STAT3 , NFKB1 , CCL4 while stimulated the transcription of gene repressed in psoriasis, IL-1RN in peripheral blood cells or lesional skin [ 75 ].…”
