Genetic tools: the use of biochemical and molecular markers.

Abstract: Several types of genetic marker are available to forest conservation geneticists, including isozymes, terpenes, restriction fragment length polymorphisms (RFLPs), microsatellites, random amplified polymorphic DNAs (RAPDs) and amplification fragment length polymorphisms (AFLPs). The techniques underlying these markers are explained in terms that are accessible to those unfamiliar with molecular biology. The strengths and weaknesses of the various marker systems are elucidated via comparison. Guidelines are give… Show more

“…In spite of their ecological and economic importance, the Gohilwari goats are inadequately characterized particularly at DNA level. Microsatellites in particular are useful in conservation genetics because the high degree of polymorphism makes them extremely informative and gives them very high discriminating power [12] , allowing for a thorough assessment of genetic variation and structure within and among populations [6] . Genetic diversity is essential for the long-term survival of the species and populations because it provides the raw material for adoption and evolution, especially when environmental conditions have changed [10,29] .…”

“…Microsatellites have high levels of polymorphism (Glaubitz and Moran 2000) and are, therefore, extremely useful for fine-scale genetic analyses. As microsatellites are a relatively recent discovery, there have been only a few eucalypt population studies using them (Byrne et al 1996;Brondani et al 1998;Steane et al 2001;McGowen et al 2001).…”
