The aim of this study was to characterise the genetic diversity, genetic relationship and population structure within local Turkish Denizli chicken subpopulations in rural areas using 19 microsatellite markers. To assess the uniqueness and relations of the Denizli subpopulations we used six local Italian chicken breeds' samples, which were genotyped in the same laboratory with the same molecular markers. In total, 105 alleles were found across 19 microsatellite loci with a mean number of 5.53 alleles per locus. Considering all subpopulations and loci, genetic differentiation based on global F ST was 0.030 (p < .01). Global F IS values (0.200) indicated that non-random mating occurred in all subpopulations of Denizli fowl and all subpopulations deviated significantly (p < .01) from HWE. Over all subpopulations, the mean observed heterozygosity was 0.473, ranging from 0.399 to 0.562. Genetic differentiations between pairs of subpopulations based on the proportion of shared alleles ranged 0.140-0.297. The neighbour-net tree, based on marker estimated kinship distances, separated Denizli subpopulations and Italian breeds into two main clusters. The most likely number of different populations was estimated using the clustering procedure implemented in STRUCTURE. Structure analysis showed a clear separation of the Denizli fowl subpopulations from the Italian populations. A second step sub-clustering allowed discriminating among the six subpopulations of Denizli breed. The results of this study can be used as baseline genetic information to place breeders' flocks in conservation programmes, controlling inbreeding and safeguarding the genetic variability of the populations.

ObjectiveThis study was conducted to compare morphological defects, viability, motility (MOT), fertility (F), and hatchability (H) in four Korean native chicken breeds (KNCBs), and to evaluate whether defective segments of spermatozoa are associated with MOT, F, and H.MethodsFour KNCBs, including Korean Ogye (KO), Hwangbong (HB), Hyunin Black (HH), and Hoengseong Yakdak (HY) were used. White Leghorn (WL) was used as a control. Nine cocks from each breed were randomly assigned into three groups. Semen was collected by abdominal massage method. Eosin-nigrosin staining method was used to identify live-dead spermatozoa. Different segments and specific morphological defects of spermatozoa were identified using 4′, 6-diamidino-2-phenylidole and MitoTracker Red CMXRos. F and H rates were evaluated following artificial insemination (AI).ResultsKO had the highest MOT rate compared to HY. Viable normal sperm rates of KO and HH were high and comparable with WL. HY spermatozoa had the highest viable abnormal sperm (VAS) or morphological defect rate followed by HB. Likewise, HB spermatozoa had the highest dead sperm (dead) rate compared to KO, HY, and WL. Bent, coiled, detached, broken, and knotted were common identified specific morphological defects for all breeds. Most morphological defects were at the head and tail in all breeds. VAS showed strong negative correlation with MOT (r = −0.697) and F (r = −0.609). Similarly, defective tail was negatively correlated with MOT (r = −0.587), F (r = −0.797), and H (r = −0.448). The F and H rates of KO and WL were comparable.ConclusionThese data indicate that most identified specific morphological defects are at the head and tail. VAS and defective tail were associated with poor motility, F, and H. KNCBs showed more morphological defects than WL. Finally, these results will facilitate successful AI and semen cryopreservation.

The aim of this study was to evaluate the genetic diversity and relationships of Ogye populations in Korea. A total of 243 genomic DNA samples from 6 Ogye population (Yeonsan Ogye; YSO, Animal Genetic Resources Research Center Ogye; ARO, Chungbuk Ogye; CBO, Chungnam Ogye; CNO, Gyeongbuk Ogye; GBO, Seoul National University Ogye; SUO) and 3 introduced chicken breeds (Rhode Island Red; RIR, White Leghorn; LG, Cornish; CN) were used. Sizes of 25 microsatellite markers were decided using GeneMapper Software(v 5.0) after analyzing ABI 3130XL. A total of 153 alleles were observed and the range was 2 to 10 per each locus. The mean of expected and observed heterozygosity and PIC (Polymorphism Information Content) value was 0.53, 0.50, 0.46 respectively. The lowest genetic distance (0.073) was observed between YSO and SUO, and the highest distance (0.937) between the RIR and CBO. The results of clustering analysis suggested 3 clusters (ΔK=7.96). Excluding GBO population, 5 Ogye populations (YSO, ARO, CBO, CNO, SUO) were grouped in same cluster with high genetic uniformity (0.990, 0.979, 0.989, 0.994, 0.985 respectively). But GBO population was grouped in cluster 1 with low genetic uniformity (0.340). The results of this study can be use to basic data for the genetic evaluation and management of Ogye populations in Korea.
