Genetic characterisation of adenovirus type 8 isolated in Hiroshima city over a 15 year period

Abstract: Aims: To investigate the genetic differences among the strains of adenovirus type 8 (Ad8) circulating in Hiroshima city, Japan, and to study their circulation pattern. Methods: One hundred and twenty nine strains of adenovirus type 8 (Ad8) were isolated in Hiroshima City over a 15 year period (1983-97) from patients with keratoconjunctivitis, and analysed with six restriction enzymes-BamHI, HindIII, PstI, SacI, SalI, and SmaI-to investigate possible relations among the isolates and their genetic variability. “…Until now, 51 serotypes have been identified. Among them adenovirus type 3, 4, 7, 8, 19 and 37 have been found to be responsible for keratoconjunctivitis worldwide (1,8). Interestingly, in the present study Ad3 was detected with the rather high incidences 4.0% and 30% and Ad8 with 8.0% and 20.0% in Japan and Korea, respectively.…”

“…Many investigators have used genomic characterization by different restriction endonucleases to establish genomic variants of Ad8, and around 20 genotypes have been identified [Fujii et al, 1983[Fujii et al, , 1984Kemp et al, 1983;Wadell, 1984]. Genome typing can be also done with subgenus-or type-specific PCR primers Tanaka et al, 2000;Adhikary et al, 2003]. Recently, some methods have suggested that the combination of PCR and RFLP analysis facilitate the typing procedure [Kidd et al, 1996;Saitoh-Inagava et al, 1996].…”

“…Ad8 has a much higher tropism for conjunctival cells than Ad19 and Ad37 and produces severe clinical manifestations and pathological alterations. This type persists in the population and causes sporadic epidemics in many countries [Kemp and Heirholtzer, 1986;Chastel et al, 1988;de Jong et al, 1992;Corsaro et al, 1999;Tanaka et al, 2000;Adhikary et al, 2003]. “…Restriction fragment length polymorphism (RFLP) analysis has been commonly used to identify genotypes in the epidemiological study of HAdVs (34)(35)(36)(37)(38)(39)(40)(41)(42). This method can divide genotypes that cannot be differentiated by sequence analysis of the hexon gene (35).…”
