Gancidin W, a potential low-toxicity antimalarial agent isolated from an endophytic Streptomyces SUK10

Abstract: Endophytic Streptomyces strains are potential sources for novel bioactive molecules. In this study, the diketopiperazine gancidin W (GW) was isolated from the endophytic actinobacterial genus Streptomyces, SUK10, obtained from the bark of Shorea ovalis tree, and it was tested in vivo against Plasmodium berghei PZZ1/100. GW exhibited an inhibition rate of nearly 80% at 6.25 and 3.125 μg kg−1 body weight on day four using the 4-day suppression test method on male ICR strain mice. Comparing GW at both concentrati… Show more

“…Diketopiperazine gancidin W (GW) was isolated from the Streptomyces sp. SUK10, an endophyte from the bark of Shorea ovalis tree, and it was tested in vivo against Plasmodium berghei PZZ1/100 (Zin et al, 2017 ). Kakadumycin A, a quinoxaline antibiotic with broad spectrum activity against Gram-positive bacteria and malaria parasites was reported from endophytic Streptomyces sp.…”

“…Thus, endophytic actinomycetes associated with traditionally used medicinal plants, especially in the tropics, could be a rich source of functional metabolites (Golinska et al, 2015). In the literature, endophytic actinomycetes associated with medicinal plants have shown the ability to synthesize many valuable bioactive compounds, including anticancer, antiviral, antimicrobial, antifungal and antiparasitic agents (Inahashi et al, 2015;Tanvir et al, 2016;Zhang et al, 2014;Zin et al, 2017). A number of the secondary metabolites are novel in structure and have broad-spectrum bioactivities that could be potentially applicable in the pharmaceutical, medical and agricultural sectors (Golinska et al, 2015;Matsumoto and Takahashi, 2017).…”

“…Diketopiperazines are natural cyclic dipeptides produced by microorganisms with a broad range of biological activities such as antimicrobial, anticancer, antiviral, antifungal, antibacterial, and antihyperglycemic [45,46]. Likewise, cyclo(L-Leu-L-Pro) cyclodipeptide (6), a diketopiperazine known as gancidin W, has been reported as potent antimalarial and antimicrobial agent with MIC values between 0.25 and 12.5 μg/mL against Enterococcus faecalis, Escherichia coli, Staphylococcus aureus, Micrococcus luteus, Candida albicans, and Cryptococcus neoformans [46,47].…”
