Introduction Benign prostatic hyperplasia (BPH) is a major chronic disease affecting men, and the therapeutic agents currently used to manage it have significant side effects. As a result, an alternative medicine with improved therapeutic properties with no side effects is desperately needed. The current investigation aims to study whether the Caesalpinia bonduc seed extracts (ethanolic-A, hydroalcoholic-B, and aqueous-C) have inhibitory potential on testosterone propionate (TP)-induced BPH in Wistar rats. Methods Wistar rats (male) were randomly allocated to one of five groups: control, BPH (TP-3 mg/kg, subcutaneously daily), low dose (TP + C. bonduc seed extracts – 200 mg/kg body weight), high dose (TP + C. bonduc seed extracts – 400 mg/kg body weight), and standard drug (TP + finasteride – 10 mg/kg body weight). At the end of drug treatment, the rats were sacrificed and their serum and prostates were taken for biochemical and histological studies. Results C. bonduc seed extracts treatment significantly decreased prostate weight and prostatic index in rats with TP-induced BPH. The seed extracts exhibited a potent inhibitory effect on dihydrotestosterone (DHT) in serum and prostate. In addition, the PSA level in the serum showed a noteworthy decrease in comparison with the BPH group. Histopathological examination also indicated that extracts improved the tissue morphology of the prostate significantly. Out of three extracts tested, ethanolic and hydroalcoholic extract recorded significant effect. Finally, liquid chromatography quadrupole time-of-flight mass spectrometry (LC/MS-QTOF) analysis showed that the major compounds present in the extracts were tocopherols, fucosterol, linoleic acid, β-amyrin, β-sitosterol, campesterol, cassane furanoditerpene, norcassane furanoditerpene and other diterpenes. Conclusion Thus, C. bonduc seed extracts could be a potential source for the formulation of new drug for managing BPH. To the best of our knowledge, this is the first scientific animal investigation into the use of C. bonduc seed extract for the management of BPH.

Silk fibroin from Bombyx mori has traditionally been utilized for garment production; however, recently, it has become feasible to manufacture edible and cosmetic products using this material. Gamma‐ray irradiation can improve physiological activity through the modification of the structure of proteins in biological materials. In this study, we examined whether gamma ray‐irradiated fibroin protein (gFP) has a protective effect on tumor necrosis factor alpha (TNF‐α)‐induced cellular stress through a change in protein structure. First, we confirmed that the structure of FP was changed by gamma‐ray irradiation using electrophoresis, UV spectra and Fourier‐transform infrared spectroscopy (FT‐IR). We further investigated the cytoprotective potential of 20 kGy‐irradiated fibroin protein (gFP20) in human skin keratinocytes (HaCaT cells) exposed to extracellular stress. gFP20 effectively decreased TNF‐α‐induced matrix metalloproteinase‐1 (MMP‐1) overexpression and restored reduced type‐1 procollagen in HaCaT cells. This reduction occurred concomitantly with suppressed nuclear factor‐κB (NF‐κB) nuclear translocation, resulting in a decreased level of inflammatory mediator and pro‐inflammatory cytokines. Furthermore, gFP20 protected cells from TNF‐α‐induced oxidative stress by attenuating reactive oxygen species (ROS) overexpression and increasing the production of the antioxidant enzyme heme oxygenase‐1 (HO‐1) through the stimulation of the translocation of nuclear factor erythroid 2‐related factor 2 (Nrf2) into the nucleus. Taken together, our findings indicate that low‐dose irradiated fibroin protein (gFP20) could be considered as a functional material for skincare products.

Recently, as the anti-aging role of melanin in the skin and the inhibition of melanin production has been identified, the development of materials capable of maintaining skin homeostasis has been attracting attention. In this study, we further investigated the anti-melanogenic effect of Codonopsis pilosula extract (CPE) and, under oxidative stress, the cytoprotective effect in Melan-a melanocytes exposed to H2O2. First, CPE treatment significantly reduced melanin production by inhibiting melanogenesis-associated proteins, including microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase-related protein 2 (TRP 2), as a result of the phosphorylation of MAPK/JNK in Melan-a cells. Next, to investigate the protective effects of the CPE on oxidative-stress-induced skin injury and its molecular mechanism, we determined the effect of CPE after inducing oxidative stress by exposing melanocytes to H2O2. CPE protected cells from H2O2-induced cytotoxicity by reducing the expression of the gene encoding the Bax pro-apoptotic protein, whereas it induced the genes encoding the B-cell lymphoma 3 (Bcl2) family and MITF, which is a transcriptional regulator that promotes melanocyte differentiation. Furthermore, our results show that CPE enhanced the production of autophagy-related proteins such as Beclin-1 and light chain 3 (LC3) II; this was substantially reversed by 3-methyladenin (MA, an autophagy inhibitor) pretreatment. Collectively, our findings demonstrate that CPE treatment exhibits not only an anti-melanogenic effect in normal melanocytes, but also a cytoprotective effect in melanocytes subjected to oxidative stress by inducing autophagy and MITF expression. Therefore, we believe that CPE is a potent candidate for cell maintenance in melanocytes.
