Frequency of sister-chromatid exchange among greenhouse farmers exposed to pesticides

SUMMARYMicronucleus (MN) is a biomarker widely used in biomonitoring studies for determining the genetic risk associated with exposure to pesticides. The purpose of this study was to assess damage to the genetic material of workers occupationally exposed to pesticides as detected in micronucleus tests. The research included 119 subjects divided into three groups: a control group of 39 subjects, a group of 40 subjects exposed to pesticides as producers and a group of 40 pesticide applicators in the field. A Mann-Whitney U-test displayed statistically significant differences between the parameter means of all variables, and the control group. Significant differences were observed between males involved in pesticide production and application for the parameter MN4, then between non-smoking producers and applicators regarding parameters MN2, MN3, MN4 and NB, as well as between the control and applicator groups for parameter MN2, and between producers and applicators for parameter MN3. Spearman's correlation test showed a positive correlation between the frequency of micronuclei and age of respondents, as well as their smoking habits. A statistically significant difference in relation to cytogenetic parameters was detected between the respondents working in pesticide production and those working in the field. The results suggest that applicators in the field do not use adequate personal protective equipment. Regular biological monitoring of workers exposed to pesticides is required.

Pesticides are one of the most potent environmental contaminants, which accumulate in biotic and abiotic components of ecosystems. Acetamiprid (Acm), a neonicotinoid insecticide, and Propineb (Pro), a dithiocarbamate fungicide, are widely used to control sucking insects and fungal infections on crops, respectively. The present study was undertaken to investigate the genotoxic effects of these compounds, individually and in mixtures, in mouse germ cells by using the sperm morphology assay. Mice were injected intraperitoneally with 0.625, 1.25, and 2.50 µg mL -1 of Acm, 12.5, 25, and 50 µg mL -1 of Pro, and their mixture at the same concentrations over 24 and 48 h. Acm did not significantly increase the percentage of abnormal sperm at any concentration. The frequency of abnormal sperm significantly increased after 24 and 48 h of exposure to 50 µg mL -1 of Pro. The mixtures of 2.50 µg mL -1 of Acm and 50 µg mL -1 of Pro induced sperm abnormalities antagonistically both after 24 and 48 h of exposure. Results suggest that Acm was non-genotoxic for mouse germ cells, while Pro may have been a germ cell mutagen due to the observed increase in the frequency of sperm abnormalities. However, to gain better insight into the mutagenicity and DNA damaging potential of both of these pesticides, further studies at molecular level should be done.

Background: Sister chromatid exchange (SCE) in human peripheral blood lymphocytes is one of the most extensively studied biomarkers employed to evaluate genetic damage subsequent to pesticide exposure. Objective: To estimate the pooled levels of SCE in human peripheral blood lymphocytes among population exposed to pesticide. Materials and Methods: Meta-analysis on the association between SCE frequency and pesticide exposure was performed with STATA 10.0 software package and Review Manager 5.0.24 in this study. Results: The overall means of SCE were 7.88 [95% confidence intervals (95%CI): 6.71-9.04] for exposure group and 6.05 (95%CI: 5.13-6.95) for controls, respectively. There was statistically significant difference in the SCE frequency in human peripheral blood lymphocytes between pesticide-exposed groups and control groups, and the summary estimate of weighted mean difference was 1.69 (95%CI: 1.01-2.38). We also observed that pesticide-exposed population had significantly higher SCE frequency than control groups among smokers, nonsmokers, pesticide applicator, pesticide producer, other exposure population and Asian population in stratified analyses. Conclusions: Data indicate that the SCE frequency in human peripheral blood lymphocytes might be an indicator of early genetic effects for pesticide-exposed populations. s Keywords: Sister chromatid exchange -pesticide -peripheral blood lymphocytes -meta-analysis
