Free Amino Acids in Plasma and Tissue of Rats Born to Underfed Dams

Abstract: The importance of prenatal and postnatal nutrition on the development of the progeny has been well documented. Severe food restriction results in infertility in women (1). Maternal malnutrition in rats retards DNA replication and the synthesis of RNA and protein (2-4). Results from our laboratory have shown that underfeeding of the dams during gestation and lactation had an adverse effect on the growth rate of the offspring (5). These stunted animals required more feed for growth and excreted more urinary nitr… Show more

“…Sulfur amino acids and taurinc arc well differenciated with this method, obtaining values comparable with the ones described in the literature rt6, 23,26] but somewhat higher than in other papers [17,20,22,25] ; our maximum is in form of taurine as in elsewhere r16]. The other amino acids, mainly essential ones, present values very much close to the ones described with the exception of tyrosine, that is constantly found in a level somewhat higher and Trp, whose [28] values are more in accordance with the « total » Trp in human plasma than with the values indicated in the literature for rats.…”

“…It is interesting to point out that in quantitative terms the most important amino acids found were the gluconeogenetic ones (Glx, Ala, Ser, Pro) [17,21,23,24,25] together with the branched chain ones (Leu, Ile and Val\ r16, 18, 26, 27]. It was found that the amino acid proline was present in considerable amounts, comparable to the ones found by several authors [19,21,22] but higher [23,26,27] or lower [17] than those found recently by others. This amino acid, together with the itHyp is perfectly quantified with…”

“…Plasma samples were mixed with two volumes of 5% (w/w) trichloroacetic acid, and centrifuged (4°C, 20 min, 8,000 g ) immediately after blood collection to remove precipitated protein. To prepare deproteinized tissue extracts, tissues were homogenized by using 5% trichloroacetic acid as described previously [21]. All samples were kept at 4°C during all steps to minimize chemical reactions of thiol metabolites.…”

“…A deproteinized tissue extract was prepared by homogenizing a portion of the cerebral hemispheres with 5% trichloroacetic acid [51]. After centrifugation (4°C, 20 min, and 8000 g) the clear supernatant layer was decanted and washed several times with ether to remove any trace of lipids.…”
