Flavonoids from Whole-Grain Oat Alleviated High-Fat Diet-Induced Hyperlipidemia via Regulating Bile Acid Metabolism and Gut Microbiota in Mice

Abstract: A high-fat diet (HFD) causes hyperlipidemia, which worsens disturbances in bile acid (BA) metabolism and gut microbiota. This study aimed to investigate the regulation of flavonoids from whole-grain oat (FO) on BA metabolism and gut microbiota in HFD-induced hyperlipidemic mice. The experiment results showed that FO improved serum lipid profiles and decreased body weight and lipid deposition in HFD-fed mice. Through real-time qualitative polymerase chain reaction (RT-qPCR) and Western blot assays, by up-regula… Show more

“…In this study, negative correlation between Lachnospiraceae_ND3007_Group with BMI and fat deposition was identified for the first time. In previous reports, Colidextribacter and Alloprevotella were positively correlated with hyperlipidemia in rats and daily gain in pigs, respectively [ 49 , 50 ], and we found them in the cecum of high-BMI Hu sheep. In particular, Saccharivibrio belonging to Proteobacteria presented high relative abundance in the cecum of high-BMI Hu sheep.…”

“…In addition, the mRNA expressions of key genes including cholesterol-7α-hydroxylase ( CYP7A1 ), bile salt export pump ( BSEP ) and Na+/taurocholate cotransporting polypeptide ( NTCP ) related to bile acid biosynthesis and metabolism were analyzed ( Duan et al, 2021 ). Compared with the Control group, the transcription levels of CYP7A1 , BSEP and NTCP were obviously down-regulated in mice with excessive alcohol intake (the Model group) ( p < 0.05).…”

“…Additionally, the fundamental effect of P. gingivalis on the gut microbiota was evident after the change in diet (TP3 in Figure 4B ). Although the role of these bacteria in liver pathology has not been clarified, Colidextribacter has recently been reported to be positively correlated with serum levels of oxidative markers ( 50 ) and hyperlipidemia-related parameters ( 51 ). In addition, functional enrichment analysis of the expression profiles of genes in the liver demonstrated that the genes involved in the response to oxidative stress were enriched (Cluster 4) in Pg mice.…”
