This work was designed to investigate biological impacts at 3 dates (day 0, day 7 and day 21) on black scallops (Mimachlamys varia) in the three ports areas of La Rochelle town in winter 2017. In order to assess the biological effects on the wild population of black scallops, bivalves were place in four different locations: in the three ports (semi-closed areas), and in a marshland uncontaminated site (closed area). Biomarkers of effects (heavy metals) and exposure (oxidative stress and immunological effects) were assessed in the digestive glands of specimens in order to compare two techniques of sampling: “pool” technique and “inter-subject” technique. Our findings reported in the both techniques show significant modulation of GST (detoxification), SOD (antioxidant response) and MDA (lipid peroxidation) in bivalves exposed to a specific contamination in each port. Laccase-type enzyme also highlighted an important aspect in terms of biomarker response of the immune function at the 7th day of exposition. Overall, our study demonstrated that the “pool” technique using the same quality indicator M. varia could be used to obtain reliable results at lower costs. In contrast, in fundamental context, the “inter-subject” technique could bring more precise results to light. However, it requires burdensome and costly handling.

Ports are a good example of how coastal environments, gathering a set of diverse ecosystems, are subjected to pollution factors coming from human activities both on land and at sea. Among them, trace element as copper represents a major factor. Abundant in port ecosystem, copper is transported by runoff water and results from diverse port features (corrosion of structures, fuel, anti-fouling products, etc.). The variegated scallop Mimachlamys varia is common in the Atlantic port areas and is likely to be directly influenced by copper pollution, due to its sessile and filtering lifestyle. Thus, the aim of the present study is to investigate the disruption of the variegated scallop metabolism, under a short exposure (48 h) to a copper concentration frequently encountered in the waters of the largest marina in Europe (82 μg/L). For this, we chose a non-targeted metabolomic approach using ultra-high performance liquid chromatography coupled to high resolution mass spectrometry (UHPLC-HRMS), offering a high level of sensitivity and allowing the study without a priori of the entire metabolome. We described 28 metabolites clearly modulated by copper. They reflected the action of copper on several biological functions such as osmoregulation, oxidative stress, reproduction and energy metabolism.

BackgroundScallops (Bivalvia: Pectinidae) present extraordinary variance in both mitochondrial genome size, structure and content, even when compared to the extreme diversity documented within Mollusca and Bivalvia. In pectinids, mitogenome rearrangements involve protein coding and rRNA genes along with tRNAs, and different genome organization patterns can be observed even at the level of Tribes. Existing pectinid phylogenies fail to resolve some relationships in the family, Chlamydinae being an especially problematic group.ResultsIn our study, we sequenced, annotated and characterized the mitochondrial genome of a member of Chlamydinae, Mimachlamys varia—a species of commercial interest and an effective bioindicator—revealing yet another novel gene arrangement in the Pectinidae. The phylogeny based on all mitochondrial protein coding and rRNA genes suggests the paraphyly of the Mimachlamys genus, further commending the taxonomic revision of the classification within the Chlamydinae subfamily. At the scale of the Pectinidae, we found that 15 sequence blocks are involved in mitogenome rearrangements, which behave as separate units.ConclusionsOur study reveals incongruities between phylogenies based on mitochondrial protein-coding versus rRNA genes within the Pectinidae, suggesting that locus sampling affects phylogenetic inference at the scale of the family. We also conclude that the available taxon sampling does not allow for understanding of the mechanisms responsible for the high variability of mitogenome architecture observed in the Pectinidae, and that unraveling these processes will require denser taxon sampling.
