Final Report on the Amended Safety Assessment of Propyl Gallate1

Abstract: Propyl Gallate is the n-propyl ester of gallic acid (3,4,5-trihydroxybenzoic acid). It is soluble in ethanol, ethyl ether, oil, lard, and aqueous solutions of polyethylene glycol (PEG) ethers of cetyl alcohol, but only slightly soluble in water. Propyl Gallate currently is used as an antioxidant in a reported 167 cosmetic products at maximum concentrations of 0.1%. Propyl Gallate is a generally recognized as safe (GRAS) antioxidant to protect fats, oils, and fat-containing food from rancidity that results from… Show more

“…Its biological activities are not limited to the free-radical scavenging ability as it exhibits anti-microbial, anti-nociceptive activity, ultraviolet (UV) radiation protection, anti-cariogenesis, anti-mutagenesis, and anti-carcinogenesis effects. However, in cosmetic formulations, its concentration is low (up to 0.1 %) due to indications for skin irritation or sensitization (CIR 2007 ). Applications of propyl gallate expand into the food, pharmaceutical, adhesive, lubricant, and biodiesel industry where it has been used as an anti-oxidant additive, for more than 20 years (Zhang 2015 ).…”

“…Propyl gallate (PG) is one of the most well-known synthetic phenolic antioxidants commonly used in food processing to scavenge free radicals and block lipid peroxidation. , In the Code of Federal Regulations (CFR), PG was listed as a “generally recognized as safe” (GRAS) substance (21CFR 184.1660) and could be used in fat and oils, baked goods, meat products, snack foods, nut products, etc . The European Food Additives Directive stipulates that the maximum allowable dose of PG for fat and oils used in the specialized production of heat-treated foods is 200 mg/kg .…”

“…Propyl gallate (n-propyl gallate, n-propyl 3,4,5-trihydroxybenzoate, PG, compound III, CAS Registry number: 121-79-9) is an important synthetic substance widely used in cosmetics, foods, pharmaceuticals, and some other fields [1][2][3]. It is used as an effective antioxidant in cosmetics to stabilize vitamins, essential oils, perfumes, as well as fats and oils [1]. In foods, PG has been employed as an additive (E310) since 1948 to protect fats, oils, and fat-containing food from peroxide-induced rancidity [1].…”
