a b s t r a c tBackground: Oral cancer is the sixth most common malignancy worldwide and is thus an important cause of death and morbidity. Although several drugs have been developed for the treatment of oral cancer, naturally occurring phytochemicals have been recommended by nutritionists and health experts to treat various types of cancer. Pergularia daemia is a well-known herb distributed across India, finding many uses in medicine. Aim: The present study was mainly designed to determine the dose-response effect of the whole plant extract of P. daemia on experimental oral carcinogenesis. Materials and methods: Male golden Syrian hamsters were painted with 0.5% of 7,12dimethylbenz[a]anthracene (DMBA) in liquid paraffin on the left buccal pouch to induce oral cancer, thrice a week for 14 weeks. The anticancer effect of the ethyl acetate and methanolic extracts of P. daemia was observed to be dose dependent (100, 200, and 300 mg/kg body weight (bwt)) by assessing the bwt, tumor incidence, changes in oxidant/antioxidant levels, and histological alterations of control and experimental animals. Results: Our results showed that of the three doses used, the high dose (300 mg/kg bwt) exhibited excellent antioxidant activity in comparison to the other two doses. Moreover, our findings suggest that the methanolic extract significantly inhibited cancer development to a greater extent than the ethyl acetate extract. Conclusion: Thus, it could be concluded that the whole plant of P. daemia presents an easily accessible source of natural antioxidants, and regular consumption can reduce the incidence of oral cancer.

Receptor for advanced glycation end-products (RAGE) is a multiligand binding and single-pass transmembrane protein taken in diverse chronic inflammatory conditions. RAGE behaves as a pattern recognition receptor, which binds and is engaged in the cellular response to a variety of damage-associated molecular pattern molecules, as well as HMGB1, S100 proteins, and AGEs (advanced glycation end-products). The RAGE activation turns out to a formation of numerous intracellular signaling mechanisms, resulting in the progression and prolongation of colorectal carcinoma (CRC). The RAGE expression correlates well with the survival of colon cancer cells. RAGE is involved in the tumorigenesis, which increases and develops well in the stressed tumor microenvironment. In this review, we summarized downstream signaling cascade activated by the multiligand activation of RAGE, as well as RAGE ligands and their sources, clinical studies, and tumor markers related to RAGE particularly in the inflammatory tumor microenvironment in CRC. Furthermore, the role of RAGE signaling pathway in CRC patients with diabetic mellitus is investigated. RAGE has been reported to drive assorted signaling pathways, including activator protein 1, nuclear factor-κB, signal transducer and activator of transcription 3, SMAD family member 4 (Smad4), mitogen-activated protein kinases, mammalian target of rapamycin, phosphoinositide 3-kinases, reticular activating system, Wnt/β-catenin pathway, and Glycogen synthase kinase 3β, and even microRNAs.

Heliotropium strigosum plant is widely used in traditional medicines for treatment of various ailments. In current study, an effort was made to evaluate phenolic profile as well as antidiabetic and antioxidant activities of crude methanol extract and different solvent (n-hexane, ethyl acetate, chloroform and aqueous) fractions of H. strigosum. Total phenolic contents were determined by spectrophotometric assay. Polyphenolic compounds in crude methanol extract and each solvent fraction were identified by reverse phase high performance liquid chromatography (RP-HPLC). Antioxidant and antidiabetic activities were determined using DPPH and α-amylase inhibition assays, respectively. The outcome of spectrophotometric assay showed that methanol extract had higher amount of total phenolic (84.50 ± 0.06 µg GAE/ mg of plant extract) contents than aqueous, ethyl acetate, chloroform and n-hexane fractions, respectively. The RP-HPLC analysis revealed the maximum number of phenolic (chromotropic acid, quercetin, trans-4-hydroxy-3-methoxy cinnamic acid, vanillic acid, gallic acid, caffeic acid, m-coumaric acid, p-coumaric acid, syringic acid, sinapic acid and ferulic acid) compounds in methanol extract. Methanol extract (IC 50 = 8.97 µg/mL) exhibited the maximum antidiabetic activity followed by aqueous (IC 50 = 20.04 µg/mL), ethyl acetate (IC 50 = 27.79 µg/mL), chloroform (IC 50 = 56.87 µg/mL) and hexane (IC 50 = 32.16 µg/mL) fractions, respectively. The outcome of antioxidant assay revealed that methanol extract was the leading one with regard to antioxidant activity at different doses (10 to 250 µg/mL). The current study concludes that H. strigosum solvents extracts with significant phenolic profile and potent biological activities could be explored for potential uses in neutraceutical and pharmaceutical industries.
