Evidence for involvement of Dicer‐like, Argonaute and histone deacetylase proteins in gene silencing in Phytophthora infestans

Abstract: Gene silencing may have a direct or indirect impact on many biological processes in eukaryotic cells, and is a useful tool for the determination of the roles of specific genes. In this article, we report silencing in Phytophthora infestans, an oomycete pathogen of potato and tomato. Gene silencing is known to occur in P. infestans, but its genetic basis has yet to be determined. Genes encoding the major components of the RNA interference (RNAi) pathway, Dicer-like (Pidcl1), Argonaute (Piago1-5) and RNA-directe… Show more

“…The sorghum SbACTIN (Sb01g010030) and 18S rRNA genes were used for normalization of expression levels (Yang et al 2004;Shen et al 2010). All calculations and statistical analyses were performed as described in the ABI PRISM 7700 Sequence Detection System User Bulletin #2 (Applied Biosystems, USA) slightly modiWed by Vetukuri et al (2011).…”

“…RNA silencing pathways in these unicellular eukaryotes have revealed some striking differences to several model organisms, such as atypical DCL proteins and unusual biogenesis of miRNAs [25], [26]. To advance the understanding of gene silencing processes operating in P. infestans , we have previously shown that P. infestans possesses the components of canonical gene silencing pathways similar to those of other eukaryotes [27]. However, genes encoding silencing-related proteins such as cytosine methyltransferases, RNA polymerase IV, Drosha and ERI1, present in other organisms, were not identified from its genome sequence.…”

“…Previously, knowledge of epigenetic mechanisms in oomycetes was not available, except a few indirect evidences suggesting that histone acetylation may be involved in gene expression [8, 9, 74]. To our knowledge, this is the first study to identify and analyze the evolution and the express pattern of HATs and HDACs in oomycetes.…”
