Two loci in sorghum with NB-LRR encoding genes confer resistance to Colletotrichum sublineolum

Abstract: The aim of this work was to identify plant resistance genes to the sorghum anthracnose fungus Colletotrichum sublineolum. cDNA-AFLP transcript profiling on two contrasting sorghum genotypes inoculated with C. sublineolum generated about 3,000 informative fragments. In a final set of 126 sequenced genes, 15 were identified as biotic stress related. Seven of the plant-derived genes were selected for functional analysis using a Brome mosaic virus-based virus-induced gene silencing (VIGS) system followed by fungal… Show more

“…Our result is consistent with three previous studies in sorghum that reported NBS-LRR genes conferring resistance to fungal pathogens; St-R gene cluster conferring resistance to Setosphaeria turcica causing northern leaf blight in maize [55], Cs1 and Cs2 genes conferring resistance to Colletotrichum sublineolum causing anthracnose [56] and Rph 1-2 conferring resistance to leaf rust [3,57]. …”

“…These include calmodulin (Pavir.Hb00190) [70, 71], WD domain (Pavir.J03445) [72, 73], and transposable elements (Pavir.Fb0150) [74, 75]. This highlights the diversity and potential for gene diversification of RGHs encoded by the switchgrass genome.…”

“…The barley stripe mosaic virus (BSMV)-based VIGS vector was used to characterize multiple genes for their roles in disease resistance in wheat and barley (Hein et al, 2005; Scofield et al, 2005; Zhou et al, 2007; Sindhu et al, 2008). Several scientists have developed a brome mosaic virus (BMV)-based VIGS vector, and this vector was demonstrated to be a versatile tool for rapid gene function analysis in barley, rice, and maize (Ding et al, 2006; Pacak et al, 2010; van der Linde et al, 2011; Biruma et al, 2012). In rice seedlings, 19 and 13 NAC genes were up-regulated after RSV and RTSV infection, respectively, at different days after inoculation (Nuruzzaman et al, 2010).…”
