a b s t r a c t a r t i c l e i n f oArticle history: Received 9 January 2016 Received in revised form 26 July 2016 Accepted 29 July 2016 Available online xxxx An integrative biomarker approach was employed to evaluate the environmental quality of the north coast of Shandong Peninsula along the southern Bohai Sea of China, where pollution is an imminent threat due to rapid urbanization and industrialization. A battery of biomarkers and the metal bioaccumulation in tissues of native oyster Crassostrea gigas were measured under field conditions. Integrative biomarker index (IBR) and metal body burden were calculated to differentiate the pollution status of seven sampling sites. According to our results, Xinzhuang (XZ) site was the most severely contaminated, with the highest IBR value of 3.58, while the lowest IBR value (0.04) was obtained at Penglai (PL). Such an integrated biomarker approach was proved as a useful method for environmental quality assessment in the study area.

ResumenEl presente trabajo describe la estandarización de las técnicas del Test de Micronúcleo (MN) y Ensayo Cometa para la evaluación del daño en el DNA de bivalvos marinos. La importancia de estas técnicas radica en que permiten evaluar la respuesta temprana al estrés ocasionado por agentes contaminantes y cambios ambientales. Tejidos branquiales de Semimytilus algosus y Aulacomya ater fueron utilizados para tratamientos in vivo e in vitro. Los tejidos fueron expuestos a los agentes mutagénicos Mitomicina C en concentraciones de 0,02; 0,04 y 0,06 x 10 -6 M y H 2 O 2 en una concentración de 100 μM con tiempos de evaluación de 1, 3, 6, 24 y 48 h. Diferentes soluciones para la colecta del tejido y aislamiento celular fueron evaluados obteniendo una mayor viabilidad celular en la solución salina CMFS, pH 7,3; en frío. El Test de Micronúcleo se modificó en la forma de obtener el disgregado celular, realizando la hipotonización en Citrato de sodio 0,9%, fijación en metanol por segundos posterior al frotis y la tinción con Giemsa 2%. La estandarización del Ensayo Cometa Alcalino, descrita por Wilson et al. (1998), se realizó modificando la suspensión celular en agarosa LMP (1% en solución Kenny, pH 7,5), la exposición a solución de lisis (pH 10 con DMSO y Tritón) con variaciones en el tiempo de 1 a 16 h, y la electroforesis en solución alcalina pH 13,7 y tinción con Bromuro de etidio y Nitrato de plata para geles de agarosa. Los cambios realizados en ambas técnicas permitieron obtener un alto número celular con morfología definida, observándose macrolesiones como la formación de MN y aberraciones nucleares, y la determinación cualitativa de microlesiones observadas por fragmentación y migración del DNA. These techniques are important because they can evaluate the early stress response caused by pollutant agents and environmental changes. Branchial tissues of Semimytilus algosus and Aulacomya ater were used for in vivo and in vitro treatments. Tissues were exposed to the mutagenetic agents Mitomicine C in concentrations of 0,02; 0,04 and 0,06 x 10 -6 M and H 2 O 2 in concentration of 100 μM evaluated at 1, 3, 6, 24 and 48 h. Tamanskij Bay opens into the Kerch Strait which connects the Black and the Azov seas and houses the shallow ecosystem which is the most important in the Russian part of the Black Sea. This ecosystem is dominated by sea grasses (Zostera marina etc). We have been studying bottom sediment hydrocarbons distribution and biomarkers of the pollution in Mytilus galloprovincialis for two years after the wreck of "Volgoneft' 139" and spilling of more than 1.5 thousand t of residual oil in the Kerch Strait in November 2007. The high level of hydrocarbons in bottom sediments was observed within the analysis of samples taken in 2008 (to 1.5 mg/g). The hydrocarbon level in bottom sediments significantly decreased by July 2009. The n-alkanes distribution analysis reveals two following pollution sources in study area: accidental pollution by residual oil after spill and local pollution sources related to the settlements and the ports. The study of biomarkers in mussels indicated the oil spill pollution effects only.As the considered study reveals, at present the ecosystem overcomes the negative effects of the oil spill due to its selfpurification sources. However, in case of the recurrent pollution, the situation may change.
