The current study aimed to qualitatively and quantitatively determine the phytochemical components of Cycas pectinata methanol extract (MECP), along with its antioxidant, anti-inflammatory, thrombolytic, locomotor, anxiolytic, analgesic, and antidiarrheal activities. The in vitro antioxidant activity was evaluated by DPPH scavenging assay and the total phenol and total flavonoid contents, while the anti-inflammatory activity was evaluated by a protein denaturation assay. The in vivo locomotor effects were examined using the open field test and holecross test. The anxiolytic effect was examined using the elevated plus maze (EPM) test, hole-board test (HBT), and light-dark test (LDT), while the analgesic activity was investigated using the acetic acid-induced writhing test. The antidiarrheal effect was evaluated by castor oil-induced diarrhea and gastrointestinal motility. Ten bioactive compounds were selected on the basis of their biological activities and further investigated using in silico molecular docking simulation to correlate with the identified pharmacological properties. Additionally, the ADME properties of the compounds were evaluated according to their drug-likeness profile. MECP had a maximum total phenol content of 209.85 AE 3.40 gallic acid equivalents/g extract and a total flavonoid content of 105.17 AE 3.45 quercetin equivalents/g extract, with an IC 50 value of 631.44 μg/mL. MECP (62.5-500 μg/mL) elicited 20.96-38.12% decreased protein denaturation compared to diclofenac sodium (65.40-83.50%), while a 35.72% (P < 0.001) clot lysis activity was observed for the 10 mg/mL concentration. MECP induced a dose-dependent reduction in locomotor activity, with a significant anxiolytic effect. In the analgesic test, MECP (200, 400 mg/ kg) showed a 45.12% and 58.82% inhibition in analgesia, and the 400 mg/kg dose elicited a 27.5% inhibition in intestinal motility. These findings suggest that MECP might be effective in treating antioxidant, antiinflammatory, and neuropharmacological defects, but this requires further study.

Lichens are known for their extraordinary secondary metabolites and thrombolytic activity. Parmotrema perlatum is commonly called Canary moss or Mangalore spices. This lichen is used as tspices in the food in the Indian tradition. The use of P. perlatum in medicine is based on the fact that they contain unique and varied biologically active substances, as natural Antioxidant, Anti-microbial, and Anti-coagulant. Since they are natural anti-biotics, their metabolites exert a wide variety of biological actions including anti-mycotic, antiviral, anti-inflammatory, analgesic, anti-pyretic, anti-proliferative, and cytotoxic effects, they are considered as potential drugs. They contain a variety of secondary metabolites flavonoids, terpenoids, saponins . The lichens can be used as active ingredients for the preparation of drugs for their broad range of activity. The results obtained as a range of phytochemicals like terpenoids, flavonoids, glycosides, carohydrates, proteins were detected in the leaf extracts. Fresh leaves of Parmotrema perlatum and Vitex negundo Aqueous, Chloroform, and Methanol extracts were prepared and used to find out the anti-inflammatory activity of Parmotrema perlatum and Vitex negundo by In Vitro anti-inflammatory method. The study showed significant results.

Free radicals contribute to human health problem resulting in various human diseases, including central nervous system injury, cancer, inflammations, and the decrease of organ function related to oxidation. This condition has encouraged the effort of finding new natural antioxidant and anti-inflammatory sources. Macroalgae act as excellent natural resources due to their bioactive potential with diverse applications in various fields. Eucheuma cottonii belonging to Rhodophyceae grow abundantly along Lemukutan Island waters, however, their existence has not been exploited. This study aims to evaluate antioxidant and anti-inflammatory activities of ethanol extract of E. cottonii from Lemukutan Island waters, West Kalimantan. The observation of antioxidant activity was done using the method of DPPH (1,1-diphenyl-2-picrylhydrazyl) with UV-vis spectrophotometer, while anti-inflammatory acyivity activity was determined using the RBCs membrane stability method. The ethanolic extract of E. cottonii had potential antioxidant activity with IC50 of 127.75 ppm and was classified as moderate category. Extracts showed also anti-inflammatory acyivity activity with the concentration of 219.83 ppm. Red macroalgae E. cottonii can be used as potential natural antioxidant and anti-inflammatory agent.
