The harvested plant products, specifically, the grains of cereals are major drivers of soil phosphorus (P) depletion. However, the breeding or biotechnology efforts to develop low P seeds have not been attempted because of possible adverse effects on seedling vigour and crop establishment. Several studies have contradictory observations on influence of seed P on seedling vigour. Lack of appropriate genetic material has been the major bottleneck in reaching the consensus. In this study, we used 30 EMS induced mutants of rice cultivar Nagina22 to understand the role of seed P on seedling vigour and associated physiological processes. Seedling vigour, morpho-physiological characteristics, acid phosphatases, alpha-amylase, and expression of P transporter genes were analyzed in seedlings obtained from seeds of high and low grain P mutants. The study suggests that seed P has a significant role on seedling vigour, chlorophyll content and photosynthesis process of young seedlings, and P transport from roots. Notably, we identified few mutants such as NH4791, NH4785, NH4714, NH4663, NH4614, and NH4618 which showed least influence of low seed P on seedling vigour and other metabolic processes. Therefore, these mutants can be used in breeding programs aiming for development of low P grains. Also, these and other identified mutants can be used to decipher the genetic and molecular mechanisms regulating the differential response of seed P on germination, seedling vigour and several other physiological processes influencing the crop growth and establishment.

Seed germination is the first and the most susceptible stage in plant’s growing phases, so could be considered as an index to evaluate the effect of newly developed materials like nanoparticles (NPs), providing useful information for researchers. In our experiments, germination tests have been carried out in Petri plates, containing wet filter paper and nano-primed seeds. We had biosynthesized seven nanoparticles in our previous researches, including calcinated and non-calcinated zinc oxide, zinc, magnesium oxide, silver, copper and iron nanoparticles. The effect of these biogenic nanoparticles and their counterpart metallic salts including zinc acetate, magnesium sulphate, silver nitrate, copper sulphate and iron (III) chloride was studied on two popularly grown plants, wheat and flax, in laboratory condition to obtain preliminary information for future field experiments. Germination percentage, shoot length, root length, seedlings length, root-shoot ratio, seedling vigor index (SVI), shoot length stress tolerance index (SLSI) and root length stress tolerance index (RLSI) were calculated at 2nd and 7th days of the experiment. According to the results, the response of the plants to metal containing nanoparticles and metal salts mainly depend on type of the metal, plant species, concentration of the NP suspension or salt solution, condition of the exposure and the stage of growth.

Carbon Nanoparticles Functionalized with Carboxylic Acid Improved the Germination and Seedling Vigor in Upland Boreal Forest Species

Nanopriming has been shown to significantly improve seed germination and seedling vigor in several agricultural crops. However, this approach has not been applied to native upland boreal forest species with complex seed dormancy to improve propagation. Herein, we demonstrate the effectiveness of carbon nanoparticles functionalized with carboxylic acids in resolving seed dormancy and improved the propagation of two native upland boreal forest species. Seed priming with carbon nanoparticles functionalized with carboxylic acids followed by stratification were observed to be the most effective in improving germination to 90% in green alder (Alnus viridis L.) compared to 60% in the control. Conversely, a combination of carbon nanoparticles (CNPs), especially multiwall carbon nanoparticles functionalized with carboxylic acid (MWCNT–COOH), cold stratification, mechanical scarification and hormonal priming (gibberellic acid) was effective for buffaloberry seeds (Shepherdia canadensis L.). Both concentrations (20 µg and 40 µg) of MWCNT–COOH had a higher percent germination (90%) compared to all other treatments. Furthermore, we observed the improvement in germination, seedling vigor and resolution of both embryo and seed coat dormancy in upland boreal forest species appears to be associated with the remodeling of C18:3 enriched fatty acids in the following seed membrane lipid molecular species: PC18:1/18:3, PG16:1/18:3, PE18:3/18:2, and digalactosyldiacylglycerol (DGDG18:3/18:3). These findings suggest that nanopriming may be a useful approach to resolve seed dormancy issues and improve the seed germination in non-resource upland boreal forest species ideally suited for forest reclamation following resource mining.
