Establishment and characterization of a novel murine model of pancreatic cancer cachexia

Abstract: BackgroundCachexia is a complex metabolic and behavioural syndrome lacking effective therapies. Pancreatic ductal adenocarcinoma (PDAC) is one of the most important conditions associated with cachexia, with >80% of PDAC patients suffering from the condition. To establish the cardinal features of a murine model of PDAC‐associated cachexia, we characterized the effects of implanting a pancreatic tumour cell line from a syngeneic C57BL/6 KRASG12D P53R172H Pdx‐Cre+/+ (KPC) mouse.MethodsMale and female C57BL/6 mice… Show more

“…Muscle represents more than 40% of body weight, and thus, its wasting accounts for a large part of body weight loss in cachexia. Other organs also waste, however, including cardiac muscle and adipose tissue, two organs that clearly play an important role in cachexia and pancreatic cancer outcomes . Here, we observed wasting of skeletal muscle, heart, fat, and multiple other organs, as well as activin induction in most of those organs.…”

“…Gross observation of mice under aspiration challenge showed that mice became lean with decreased physical activity. These characteristics might be similar to the muscle atrophy induced by cancer and glucocorticoids . Reduced body weight by the aspiration challenge may reflect the lean appearance of mice .…”

“…Reduced body weight by the aspiration challenge may reflect the lean appearance of mice . For physical activity, facility limitations did not allow us to apply tracking devices to quantify the locomotor activity, as previously shown . We evaluated the diaphragm as a respiratory muscle.…”

“…Again, no sexual dimorphism was observed in either genotype. It is important to note that a decrease in LMA is the first measurable behavioural change in PDAC cachexia and precedes anorexia . In addition, BT was simultaneously recorded with the same MiniMitter system.…”

“…In the present study, we utilized a murine PDAC model that reliably induces a robust and reproducible cachexia phenotype . We tested the hypothesis that MyD88 signalling plays an essential role in mediating PDAC‐induced systemic and central nervous system (CNS) inflammation, thereby serving as a central mechanistic driver of metabolic derangement and cachexia.…”
