Enhanced taxane production and secretion from Taxus baccata cell culture by adding dimethylsulfoxide

Abstract: The ability of an aprotic solvent, dimethylsulfoxide (DMSO), to induce taxane synthesis and release from cell suspension culture of Taxus baccata was examined. The results showed that applying DMSO in optimal conditions not only led to enhancement in taxane excretion from the cells but also led to improvement in taxol synthesis. Maximum yields for taxol [3.34 mg/g dry cell weight (DCW)] were achieved by adding 5% DMSO to the culture at the late-exponential phase of cell growth (day 14) and culturing for 21 day… Show more

“…As reported previously, application of permeabilizing agents in cell and hairy root cultures has been shown effective to induce the secretion of secondary metabolites . For example, the application of tween 80 led to the release of 98% licochalcone A produced by Glycyrrhiza uralensis hairy roots into the culture medium and also increased the licochalcone A production by nine‐fold .…”

“…Table 1 also shows the overall yields of three tanshinones, cryptotanshinone, tanshinones-I, and tanshinone-IIA, which were stimulated by most of the surfactant and doses. As reported previously, application of permeabilizing agents in cell and hairy root cultures has been shown effective to induce the secretion of secondary metabolites [6,10,12,19,20]. For example, the application of tween 80 led to the release of 98% licochalcone A produced by Glycyrrhiza uralensis hairy roots into the culture medium and also increased the licochalcone A production by nine-fold [11].…”

“…HPLC analysis was used to conrm the presence of various Taxane compounds in the ethanolic extract according to the literature method with some modications. 25,26 Briey, the ltered ethanolic extract was concentrated to dryness in a rotary evaporator at 75 C and the residue was dissolved in 2 mL methanol and degreased by adding 1 mL water for 4 h. The ltered methanolic extracts were analyzed by reverse phase HPLC (Sykam, Eresing, Germany) with UV detection at 227 nm. A 20 mL portion of the samples was injected on to a reversephase column (Kromasil C18, 250 mm  4.6 mm).…”
