Enhanced generation of retinal progenitor cells from human retinal pigment epithelial cells induced by amniotic fluid

Abstract: BackgroundRetinal progenitor cells are a convenient source of cell replacement therapy in retinal degenerative disorders. The purpose of this study was to evaluate the expression patterns of the homeobox genes PAX6 and CHX10 (retinal progenitor markers) during treatment of human retinal pigment epithelium (RPE) cells with amniotic fluid (AF), RPE cells harvested from neonatal cadaver globes were cultured in a mixture of DMEM and Ham's F12 supplemented with 10% FBS. At different passages, cells were trypsinized… Show more

“…hRPE cells were routinely isolated and each sample was assessed by ICC tests for RPE 65 and cytokeratin as specific markers for RPE cells. [ 20 ] hRPE cells were established in the 6 th passage of general hRPE cultures. These cells made sizeable spaces, devoid of cells, between the monolayer of hRPE cells, and gradually invaded unoccupied spaces.…”

“…The study showed that the longer PMI, the time of neurospheres formation increases ( Carter et al, 2007 ). Other research conducted on the cadavers showed that adding the amniotic fluid preparation to the in vitro RPCs medium significantly increases the generation rate of retinal progenitor cells ( Sanie-Jahromi et al, 2012 ).…”

“…[1][2][3] Through the production of different fibrotic and angiogenic factors in choroidal neovascular membranes, retinal pigment epithelial (RPE) cells contribute to choroidal neovascularization (CNV) and paracrine signaling between choriocapillaris and RPE layer. [4][5][6][7] Hypoxia is an important regulator of cell migration and angiogenesis, especially under pathologic conditions, which perform its action by upregulating hypoxiainducible factor 1 (HIF-1) and vascular endothelial growth factor A (VEGFA). [8][9][10][11] VEGFA mainly regulates angiogenesis, contributing to the migration and proliferation of vascular endothelial cells and tube formation, which increase vascular permeability in angiogenesis.…”
