Correlation between ELF–PEMF exposure and Human RPE Cell Proliferation, Apoptosis and Gene Expression

Abstract: Purpose: Emerging evidence implies that electromagnetic fields (EMFs) can negatively affect angiogenesis. In this regard, the effects of extremely low frequency pulsed electromagnetic field (ELF–PEMF) exposure on the relative expression level of angiogenic factors involved in the pathogenesis of ocular disorders were evaluated in human retinal pigment epithelial (hRPE) cells in order to investigate a noninvasive therapeutic method for patients with several ocular diseases associated with neovascularization.&#x… Show more

“…The study was approved by the Ethics Committee of Golestan University of Medical Sciences (ethics code: IR.GOUMS.REC.1399.065). Cell Culture The present study utilized an RPE cell line that had undergone prior isolation and culturing procedures in our previous investigation [22] . The cells were cultured in T-25 flasks containing a complete culture medium containing 10% fetal bovine serum (FBS) and incubated at 37°C and 5% CO 2 after thawing.…”

“…The expression of VEGF-A, PDGF-A, PDGF-B, PDGF-C, PDGF-D, and GAPDH genes in cells treated with free niosome, free AUR, and niosomal AUR were measured using SYBR Green real-time polymerase chain reaction (RT-PCR, Yekta Tajhiz Azma Inc., Tehran, Iran). Table 1 contains the list of PCR primers [22,[24][25][26][27] . Statistical Analysis The 2 -∆∆Ct method was used to calculate the relative fold changes in gene expressions.…”

“…Researches show a noteworthy elevation in the percentage of tail DNA in the testes under prolonged irradiation in comparison with other organs of rats [43]. ELF -PEMFs have a notable prohibitory function in the expression of angiogenic factors in hRPE cells as pivotal cells in eye health without cytotoxic or proliferative side effects and ELF -PEMF can be employed for therapeutic uses in ocular diseases [44].…”

“…VEGF-A (located on 6p21.1 with 26 identified transcript) is the most critical factor in the angiogenesis process, which leads to an increase in the number of capillaries. As a signal protein, VEGF-A is produced by many tissues with insufficient blood circulation and functions as a part of the oxygen and nutrition delivery system [10] . Like other family members of VEGFs, it is highly conserved and considered as part of evolutionary distance between fish and mammals [11] .…”
