Enhanced apoptosis of glioma cell lines is achieved by co-delivering FasL-GFP and TRAIL with a complex Ad5 vector

Abstract: Brain tumors (BTs) are among the most malignant forms of human cancer. Unfortunately, current treatments are often ineffective and produce severe side effects. Cytotoxic gene therapy is an alternative treatment strategy, with the potential advantages of reduced toxicity to normal brain tissue. Apoptosis-inducing ''death ligands'' Fas ligand and TNF-related apoptosis-inducing ligand (TRAIL) are genes with substantial cytotoxic activity in susceptible tumor cells. Here, we compared the effectiveness of Ad vector… Show more

“…Our initial approach was to develop tissue specificity of transgene expression, by using tissue or tumor-restricted promoters to regulate a TET-derived transcription factor (tTA) which drives transcription of the selected transgene (FasL, Apoptin, Bax or TRAIL) [1][2][3][4][5][6][7][8][9] (See Figure 1a). We successfully carried out such studies with adenoviral vectors and demonstrated restricted gene expression.…”

“…Considering that apoptosis plays a central role in regulation of pituitary tissue homeostasis, the imbalance between cell death and proliferation in favor of cell survival could result in tumor formation. One of the commonly employed strategies in experimental gene therapy for cancer is to target death receptors specifically to trigger apoptosis in tumor cells (33)(34)(35)(36).…”
