Background: T cells play a key role in the pathogenesis of chronic inflammatory enteropathy (CIE) in dogs. Cluster of differentiation 3 (CD3) antigen serves as a marker for T cells. In human medicine, Ki-67 is an indicator for cell growth but there are only a few studies in dogs with CIE.Objective: To investigate Ki-67 in relation to T cells as a marker for CIE in dogs.Animals: Eleven dogs with CIE and 6 healthy beagle controls (CO). Methods: Retrospective case-control study. Dogs were clinically assessed by the Canine Chronic Enteropathy Clinical Activity Index (CCECAI). Duodenal mucosal biopsy samples were endoscopically obtained for histopathologic examination by means of the World Small Animal Veterinary Association score. Double-labeled immunofluorescence was used to investigate colocalization of Ki-67 and CD3 in epithelium and lamina propria (LP) of villi and crypts.Results: Dogs with CIE had significantly higher clinical score (median, 5.0; interquartile range [IQR], 3-7) compared to CO (all 0; P < .001). The Ki-67/CD3 doublepositive cells were significantly increased in the LP of the crypt region of CIE dogs (0.63 cells/mm 2 ; IQR, 0-0.54) versus CO (0.08 cells/mm 2 ; IQR, 0-0.26; P = .044). A significant correlation was found between CCECAI and the Ki-67/CD3 ratio in the LP of the crypt region (r = 0.670; P = .012) in dogs with CIE.Conclusions and Clinical Importance: The Ki-67/CD3 ratio is upregulated in the LP crypt region of dogs with CIE and it correlates with clinical severity. Therefore, Ki-67/CD3 could be a useful tool for detection of CIE.

Summary Objective: The objective of this study was to correlate clinical signs and diagnostic parameters with duodenal inflammatory and architectural changes in dogs with lymphocytic-plasmacytic enteropathy. Material and methods: In a retrospective study dogs presented between 2003 and 2014 with chronic gastrointestinal signs (duration > 3 weeks) and histologic evidence of intestinal lymphocytic-plasmacytic inflammation were evaluated. Clinical signs, serum albumin, cobalamin and folic acid concentrations were recorded and a sonographic, endoscopic, histologic and cytological inflammatory score was determined. Furthermore, the presence of lacteal dilation, villus stunting, crypt lesions, epithelial integrity and increased intraepithelial lympho cytes was evaluated. Results: A total of 270 dogs were retrospectively evaluated. No significant correlation was found between clinical signs and sonographic, endoscopic or duodenal inflammatory score. Dogs with histological signs of lacteal dilation (p = 0.001) and increased intraepithelial lymphocytes (p = 0.005) had significantly higher clinical scores compared to dogs without these changes. No correlation was found between clinical score and villous stunting or crypt lesions. Hypoalbuminemia and hypocobalaminemia correlated significantly with lacteal dilation (p = 0.001, p = 0.009) and increased intraepithelial lymphocytes (p = 0.036, p = 0.018). Clinical significance: Some clinical and diagnostic parameter correlate with histopathologic features whereas others do not. Morphological features seem to be more important than the intensity of the duodenal inflammation in the assessment of the disease.

Endoscopy scoring 19 was performed based on qualitative evaluation of endoscopic mucosal appearance which included friability, granularity, erosions/ulcerations and lymphatic dilatation. Background: The use of serological markers to diagnose inflammatory bowel disease (IBD) in humans is well-established. Because of the frequency of IBD in dogs and resources required for its diagnosis with current methods, new approaches are desired.Objective: The goal is to evaluate novel serologic markers to differentiate clinical cohorts in dogs with gastrointestinal (GI) disease and assess their potential to develop a serum-based IBD diagnostic test.Animals: Seventy dogs diagnosed with biopsy-confirmed IBD, 23 dogs with non-IBD predominantly acute GI diseases, and 58 normal dogs.Methods: Prospective control study. ELISA methods were developed to detect autoantibodies to polymorphonuclear leukocytes (APMNA) and calprotectin (ACNA), antibodies against gliadins (AGA), microbial outer membrane porin C (ACA), and flagellins (AFA) isolated from diseased dogs based on clinical and histopathological scoring.Results: IBD dogs displayed a 39%-76% prevalence of seropositivity against selected serologic markers that markedly decreased to 0%-13% in non-IBD and normal dogs.ROC analysis showed statistical significance in differentiating the cohorts, with seropositivity against OmpC being the highest single performance marker. The combination of markers such as OmpC and APMNA reached specificities of 93%-99% and 79%-98% and sensitivities of 76%-97% and 66%-86% when comparing IBD versus normal cohorts and non-IBD cohorts, respectively.
