Enantioselective separation of the novel antidepressant mirtazapine and its main metabolites by CEC

Abstract: In this work, the simultaneous enantioseparation of the second-generation antidepressant drug mirtazapine and its main metabolites 8-hydroxymirtazapine and N-desmethylmirtazapine by chiral CEC is reported. The separation of all enantiomers under study was achieved employing a capillary column packed with a vancomycin-modified diol stationary phase. With the aim to optimize the separation of the three pairs of enantiomers in the same run, different experimental parameters were studied including the mobile phase… Show more

“…On the other hand, only two methods use a very different selector with a glycopeptidic structure, i.e. vancomycin [45,50]. This is in accordance with what can be observed in HPLC.…”

“…One paper has been very recently published on the enantioseparation of mirtazapine and its two main metabolites, N-desmethylmirtazapine and 8-hydroxymirtazapine, by CEC [45]. The paper describes a method based on a chiral stationary phase (CSP) obtained by binding vancomycin to a silica diol sorbent; the effect of several experimental parameters on the separation was studied: mobile phase composition (buffer concentration and pH, organic modifier type and ratio and water content), stationary phase composition and capillary temperature.…”

“…Other examples of the CEC application are illustrated on biological samples. For example, CEC based on vancomycin CSP offered effective solution not only for the enantioseparation of mirtazapine and its metabolites (8-hydroxymirtazapine and N-desmethylmirtazapine) in urine samples (pretreated by off-line SPE) but also for their reliable UV absorbance detection [89].…”

“…A vancomycin-based CSP was used by Aturki et al [31] to develop a separation method to determine simultaneously mirtazapine and two of its main metabolites. A mixture of the CSP with bare silica particles could resolve all six enantiomers, but not within one run.…”
