Ellagic acid inhibits IL-1β-induced cell adhesion molecule expression in human umbilical vein endothelial cells

Abstract: Expression of cell adhesion molecules by endothelium and the attachment of monocytes to endothelium may play a major role in atherosclerosis. Ellagic acid (EA) is a phenolic compound found in fruits and nuts including raspberries, strawberries, grapes and walnuts. Previous studies have indicated that EA possesses antioxidant activity in vitro. In the present study, we investigated the effects of EA on the formation of intracellular reactive oxygen species, the translocation of NFkB and expression of vascular c… Show more

“…24 OxLDL antagonizes the vasodilatory effect of NO, and ROS and NO antagonize each other at the level of intracellular signaling; for example, NO released from eNOS inhibits cleavage of IB and NF-B activation, which is enhanced by ROS. 36 Consistent with a previous study that revealed that ellagic acid inhibits cytokine-induced adhesion molecule expression, 17 our data showed that the antiathrogenic effects of ellagic acid are due, at least in part, to the inhibition of adhesion molecules and subsequent monocytic adherence by moderating the reduction in eNOS expression caused by oxLDL.…”

“…12 Ellagic acid effectively lowers the levels of plasma lipids, reduces oxidative stress, and inhibits apoptosis in hyperlipidemic rabbits, 13 as well as scavenges oxygen and hydroxyl radicals, and inhibits lipid peroxidation and 8-OhdG formation in vitro and in vivo. 14 Previous studies have shown that ellagic acid inhibits oxLDL-induced aortic smooth muscle cell proliferation, 15 inhibits cytokineinduced ROS generation and expression of adhesion molecules, 16,17 and prevents oxLDL-induced apoptosis 18 in endothelial cells. The underlying mechanisms, however, are still largely unclear.…”

“…However, triphalaadministered arthritic rats exhibited significant reduction in TNF-a and IL-1b levels in serum as well as in paw tissues. This anti-inflammatory property of triphala might be due to its active components like ellagic acid, gallic acid, 4-O-methylgallic acid, and bellericanin which have shown inhibitory action on TNF-a and IL-1b (Na et al, 2006;Yu et al, 2007;Zhao et al, 2008).…”
