“…Many studies have examined total microsomal cytochrome P450 content, levels of CYP enzyme activity, or CYP protein by immunoassay in bivalve mollusks exposed to chemicals (Canova et al, 1998; Livingstone, 1998; Livingstone et al, 1989; Monari et al, 2007; Monari et al, 2009; Shaw et al, 2002; Snyder et al, 2001; Sole and Livingstone, 2005; Wootton et al, 1995). The interpretation of those measurements has been complicated by low catalytic activity of microsomal enzymes with diagnostic substrates for CYP1A-like activity, such as ethoxyresorufin (ethoxyresorufin-O-deethylase activity, EROD) and benzo[a]pyrene (benzo[a]pyrene hydroxylase; BPH), and lack of specific cross-reactivity of antibodies for CYP1As, (Chaty et al, 2004; Jonsson et al, 2006a; Jonsson et al, 2006b).…”

“…ROS production also entailed lipid peroxidation and lipid metabolism disruption (Ferreira and Vale, 1998), and reproductive irregularities if germ cells are affected or the hormonal balance is perturbed (Armstrong, 1990). The correlation of DNA damage with cyp450 induction is well described in marine molluscs, like the mussels Mytilus galloprovincialis and Mytilus edulis (Canova et al, 1998;Shaw et al, 2002). These studies were conducted using heterolog mRNA probes and antibodies developed in fishes (Shaw et al, 2002).…”

These studies were conducted using heterolog mRNA probes and antibodies developed in fishes (Shaw et al, 2002). To our knowledge, no sequence for either cDNA or gene encoding Cyp450 in molluscan species was in the available databases.…”

“…The induction of this enzyme activity occurred at the same time that total PAHs was accumulated in mussels' tissues to concentrations 2.5-fold higher. Similarly, an increase of CYP1A protein up to 1.4-fold and an increased of the DNA damage (1.2-fold) occurred in mussels M. edulis transplanted from Port Quinn (reference site) to New Brighton (urban/industrial site, with high PAHs and PCBs levels) in the UK, after 13 weeks (Shaw et al 2002). Peters et al (1998) reported a preferential elevation of CYPlA-immunopositive protein with 3 weeks transplantation from a relatively clean site to a site containing 3-6 fold higher tissue levels of PAHs and PCBs is consistent with the observations of increased digestive gland CYPlA-immunopositive protein in Mytilus sp.…”

“…In recent years, many studies highlighted the importance of using an integrated approach to assess the effects of environmental changes in coastal or estuarine zones (Peters et al , 1999Shaw et al 2002). The application of the active biomonitoring (ABM) strategy, in which organisms of a clean or reference site are transplanted to the site that is being monitored, gives the advantage of assuring comparable results, reducing the variability normally found in field studies .…”
