Electron Microscopic Evaluation of Altered Collagen Structure Induced by N-Monochloroglycine (GK-101)

Abstract: The effects of N-monochloroglycine (NMG) solutions on the structure of collagen were studied by transmission electron microscopy. In addition to finding that collagen structure was altered by NMG solutions, the results offer evidence that buffer and water also exert some effect on collagen fiber structure.

“…In the 1970s, Habib et al [6] and Kronman et al [7] studied the effect of a nonspecific proteolytic agent, sodium hypochlorite (NaOCl), on carious dentine. Unfortunately, NaOCl not only dissolved necrotic organic material but also affected sound dentine [8].…”

“…2000). Presented in two components, one containing 0.1 m amino acids and pigment, the other 0.5% sodium hypochlorite, Carisolv™ is mixed to form a red gel of amino acids with electrostatic attraction to proteins in carious dentine (Kronman et al . 1977), and chloramines; potent halide disinfectants with tissue solvent action.…”

“…A system using N-monochloro-DL-2-aminobutyric acid (NMAB) was then developed as an alternative to conventional mechanical caries removal [Schutzbank et al, 1978; review by Yip et al, 1995b]. The apparent mechanism of action of NMAB is considered to involve the chlorination of partially degraded collagen in carious dentine, and the conversion of hydroxyproline to pyrrole-2-carboxylic acid which initiates disruption of the altered collagen fibres in carious dentine [Habib et al, 1975;Kronman et al, 1977]. Clinical studies have demonstrated the efficacy of the method [McCune, 1986;Tavares et al, 1988;Yip et al, 1995a, b].…”
