Efficient purification of antiproliferative polysaccharides from Hypsizigusmarmoreus with radial flow chromatography

Abstract: The increasing commercial significance of natural polysaccharides for use in medicinal products is stimulating the development of efficient and easy scale‐up techniques for polysaccharide purification. In this research, the crude polysaccharides from submerged cultivation broth of Hypsizigus marmoreus were purified using radial flow chromatography (RFC), and the antiproliferative activity of the purified fractions was evaluated in vitro. DEAE Sepharose CL‐6B was selected to be packed in the RFC column based on… Show more

“…The vast literature (more than 1600 citations in the PubMed) on the isolation, structural elucidation, and bioactivities of mushroom polysaccharides includes reports on the following additional mushroom cultivars: Agaricus blazei [ 35 ], Agaricus brasiliensis [ 36 ], Amanita ponderosa [ 37 ], oyster mushroom [ 38 ], Auricularia polytricha [ 39 ], Boletus edulis [ 40 ], Cookeina tricholoma [ 41 ], Cordyceps militaris [ 42 ], Entoloma lividoalbum [ 43 ], Gleoestereum incarnatum [ 44 ], Ganoderma lucidum [ 45 ], Grifola frondosa [ 46 , 47 ], Hohenbuehelia serotina [ 48 ], Hypsizygus marmoreus [ 49 ], Iliodiction cibarium [ 50 , 51 ], Lactarius deliciosus [ 52 ], Lentinus edodes [ 31 , 53 ], Macrolepiota dolichaula [ 54 ], Phellinus igniarius [ 55 ], Phellinus linteus [ 56 ], Phellinus pini [ 57 ], Pholiota adiposa [ 58 ], Pholiota nameko [ 59 ], Pleurotus eryngii [ 60 ], Pleurotus ostreatus [ 55 , 61 ], Termitomyces heimii [ 62 ], Tricholoma matsutake [ 63 , 64 , 65 ], Tricholoma mongolicum [ 66 ]. A journal reviewer noted that it is, however, important to determine if the isolated polysaccharides contain polyphenolic or other bioactive compounds that might affect their activities.…”

“…To test these speculations, we will in future conduct further studies under paired-feeding conditions with examination of energy expenditure. However, an anti-proliferative effect of EHM on adipocytes, possible components of which include hypsiziprenol, hypsin, or polysaccharides as demonstrated by previous studies in vivo only (7)(8)(9)(10)17) cannot be ruled out. Moreover, dietary polyphenols, such as resveratrol, were reported to increase the expression of the UCP or other obesity-related genes, including those responsible for ␤-oxidation and lipolysis, whose expressions were unchanged in both groups (data not shown) (18), and thereby to prevent obesity.…”

“…Modern scientific studies demonstrate that mushrooms are an abundant source of a wide range of bioactive natural products. Polysaccharides are the best-known mushroom-derived substances, and have various biological activities, such as antitumor, immunomodulatory, antihyperlipidemic, hepatoprotective, antioxidative and anti-fatigue effects [1][2][3][4][5] . Polysaccharides from medicinal mushrooms such as Ganoderma lucidum, G. tsugae, G. applanatum, Cordyceps sinensis, C. militaris have been studied extensively [6][7][8][9][10] , and the research for new bioactive polysaccharide has been extended to other mushrooms.…”

Hypsizygus marmoreus is a popular edible mushroom containing a variety of bioactive substances, such as polysaccharides, proteins, trace elements, essential amino acids, lectins, terpenoids, tocopherols, ascorbic acid, enzymes and dietary fiber 11 , and has been considered as a functional food in East Asia because of its antifungal, antiproliferative, immunomodulating, antioxidant and hepatoprotective activities 1,[11][12][13] . Generally, polysaccharide could be extracted from the fruiting body of mushrooms.…”
