Effects of Insulin-Like Growth Factor-I on the Expression of Atrogin-1/MAFbx in Chick Myotube Cultures

Abstract: The expression of atrogin-1/MAFbx, a muscle-specific E3 ubiquitin ligase, is increased in catabolic conditions that result in muscle atrophy. The expression of atrogin-1/MAFbx mRNA is also decreased by the insulin-like growth factor-I (IGF-I) in mammalian skeletal muscle cell cultures. This study investigated the effect of IGF-I on the expression of atrogin-1/MAFbx in chicken skeletal muscle cell cultures. Chick myotubes were incubated with IGF-I for 1, 6, or 24 h. Protein content was increased by IGF-I (100 n… Show more

“…Although it is well known that insulin and IGF-I have a regulatory effect on the expression of atrogin-1/MAFbx in mammalian skeletal muscle, their effects in chicken muscle cells are largely unknown. We previously reported that IGF-I suppresses the expression of atrogin-1/MAFbx in chick myotubes (Nakashima et al, 2017). In the present study, insulin suppressed the mRNA expression of atrogin-1/ MAFbx in chick myotubes.…”

“…Myoblasts were isolated from the thigh muscles of 13-dayold chick embryos (Nakashima et al, 2017). Briefly, the muscle tissue obtained from the embryo was digested with dispase (Gibco, Waltham, MA USA), and the cell suspension was transferred to an uncoated culture dish to allow fibroblast attachment.…”

“…Insulin-like growth factor-1 (IGF-1) is a key regulator of muscle development and metabolism in chickens. For example, IGF-1 increased cell protein content in chicken myotube (Nakashima et al, 2017) and promoted chicken myoblast proliferation (Yu et al, 2015). Tomas et al (1998) reported enhancement of growth and feed efficiency of broiler chickens receiving exogenous IGF-1.…”

“…IGF-1 and GHR are strong growth regulators and have anabolic effects on protein and carbohydrate metabolism [ 55 ]. IGF-1 is a vital regulator for the development of muscle as it amplifies cell protein content in broiler myofiber [ 56 ] and stimulates broiler myoblast proliferation. Moreover, IGF-1 stimulates embryonic skeletal muscle growth, and skeletal myogenesis, and consequently improves chicken meat production [ 57 ].…”
