Background/Aims: Osteoarthritis is a degenerative joint disease characterized by cartilage degeneration and a chondrocyte inflammatory response that induces an inflammatory environment closely linked to extracellular matrix (ECM) degradation. Ligustilide (LIG) is a major component of the herb Radix Angelicae Sinensis, with demonstrated anti-inflammatory effects. To confirm whether LIG has an equally inhibitory effect on inflammation in human osteoarthritis chondrocytes, we performed in vivo and in vitro experiments to validate the above conjectures and determine the relevant mechanisms. Methods: Quantitative realtime PCR and western blotting were performed to evaluate the expression of MMP-3, MMP-13, ADAMTS-5, iNOS, and COX-2 at both gene and protein levels. An enzyme-linked immunosorbent assay was used to evaluate the levels of other inflammatory factors (PGE2, TNF-α, and IL-6). The PI3K/AKT and nuclear factor kappa B (NF-κB) signaling pathways were also analyzed by western blotting, whereas immunofluorescence was used to assess the expression of collagen II and aggrecan. The in vitro effect of LIG was evaluated by intraperitoneal injection into a mouse osteoarthritis model induced by destabilization of the medial meniscus. Results: LIG lowered the phosphorylation levels of p65, IκBα, and IKKα/β and suppressed the IL-1β-induced expression of MMP-3, ADAMTS-5, iNOS, and COX-2 and the inflammatory factors PGE2, TNF-α, and IL-6. LIG markedly decreased IL-1β-induced degradation of collagen II and aggrecan. In vivo results showed that LIG-treated mouse cartilage showed less damage than the control group; the Osteoarthritis Research Society International (OARSI) score was also lower. LIG further reduced the thickness of the subchondral bone plate and alleviated the synovitis. Conclusion: LIG may act as a promising therapeutic agent for osteoarthritis by attenuating IL-1β-induced inflammation in chondrocytes and ECM degradation via suppression of NF-κB activation by the PI3K/AKT pathway.

Background: The study was performed to determine whether idealized anterior cruciate ligament reconstruction (IACL-R) restores normal gait features, and whether in ammatory factors are involved in the pathogenesis of post-traumatic osteoarthritis (PTOA). Methods: Fourteen mature female minipigs were allocated to a sham group (n = 7) or an IACL-R group (n = 7). Load asymmetry during gait was recorded using a pressure-sensing walkway measurement system to evaluate the gait features of the right knee joint before and after surgery. In ammatory factors (including interleukin [IL]-1α, IL-1β, IL-2, IL-6, IL-8, IL-18, tumor necrosis factor-α, and granulocytemacrophage colony-stimulating factor) in synovial uid were measured using Luminex assays before and after surgery. Cartilage integrity and the subchondral bone plate of the right knee were evaluated using histology and imaging at 3 months postoperatively.Results: Swing time and stance time returned to their preoperative values on day 31, while maximum force, contact area, peak force ,and impulse returned to their preoperative values on day 45 after the surgery in the IACL-R group (P = 0.073, 0.053, 0.107, 0.052, 0.152, and 0.059, respectively).Thus, IACL-R restored normal gait. Compared with their preoperative concentrations, all tested in ammatory factors showed signi cantly increased concentrations in the synovial uid in the IACL-R group, especially at 3, 7, and 15 days postoperatively. X-ray, computed tomography, magnetic resonance imaging, and histological data showed severe cartilage damage in the IACL-R model. Conclusion: IACL-R restored normal gait features but caused signi cant cartilage damage, indicating that signi cantly elevated in ammatory factors maybe crucial for the pathogenesis of PTOA.

Objectives: Osteoporosis is a disease described by a skeletal degradation of bone tissue domination to increase risk of fracture. Traditionally, Rusa unicolor antler from East Kalimantan is used to treat many kinds of diseases, and one of them is disease related to bone turnover. The present research aims to analyze the effects of 70% ethanol and aqueous extracts of Rusa unicolor antler from East Kalimantan on nitric oxide inhibition, osteoblast differentiation, and mineralization related to bone turnover. Materials and methods: The nitric oxide inhibition of the extracts in LPS-stimulated RAW 264.7 cells were evaluated by Griess reagent while osteoblast differentiations of extracts were evaluated by measuring alkaline phosphatase in p-nitrophenyl phosphate and their mineralization was determined using Alizarin Red Staining method. Results: The 70% ethanol and aqueous extracts inhibited cells inflammation (40% and 80%) and stimulated osteoblast differentiation (65% and 52%), respectively. In mineralization test, the aqueous extract showed two times higher effect than that of 70% ethanol extract. u n c o r r e c t e d p r o o f Conclusion: The extracts can be considered to successfully reduce expression of inflammatory markers on osteoblasts and maintain osteoblast functions.
