Effects of early postnatal sibling deprivation on anxiety and vulnerability to cocaine in offspring rats

Abstract: Our findings that early postnatal sibling deprivation influences maternal licking/grooming behavior, as well as anxiety-like behavior and vulnerability to drugs in pups that have grown to adulthood, suggests that both sibling interaction and maternal behavior, play critical roles in individual development.

“…The behavioural locomotor activity test was based on our and others' previous studies (Boudreau and Wolf, 2005;Li et al, 2008b). Briefly, rats were habituated to the behavioural testing procedure by being placed in photocell cages for at least 1 h. They then received an injection of drug/vehicle and were immediately placed in the chambers.…”

“…Neonatal sibling deprivation during the first 2-3 weeks of life attenuates the locomotor and rewarding effects of cocaine in male rats relative to controls reared in a litter of four males and four females (Li et al, 2008). Although no differences were reported in females, only one conditioning regimen was examined, and thus it is possible that sensitivity of females to stimulants may be affected under different conditioning parameters.…”

“…As described in previous studies [14,15] , 25 squares (20.0 cm×20.0 cm) were delineated on the floor, and the number of times the rats crossed between squares was counted by two observers (blind to the experimental groups) over a 10-min period. Crossing, as a measure of locomotion, was counted when the rat moved all four legs from one quadrant to another.…”
