Cytogenetic effects of Anilofos which was widely used in agriculture, was evaluated in Allium cepa root meristematic cells. In the Allium root growth inhibition test EC 50 value was determined 50 ppm and 1/29 EC 50 (25 ppm), EC 50 (50 ppm) and 2 9 EC 50 (100 ppm) concentrations of Anilofos were applied to onion roots. A negative and positive control were used in the experiment in parallel. According to results mitotic index decreased with increasing the Anilofos concentrations in all application groups and each exposure time, while disturbed anaphase-telophase, choromosome laggard(s), stickiness and anaphase bridge(s) were observed. In anaphase-telophase cells, c-metaphase, disturbed nucleus and binuclear cells were observed in other anomalies. The results were also analyzed statistically by using Dunnett t test (2-tailed) and all concentrations of Anilofos were found significant.

Genotoxic and mutagenic effects of aqueous extract from aerial parts Linaria genistifolia (L.) Mill. subsp. genistifolia, Plantaginaceae (Lg-ext) were investigated by using both Allium cepa root meristematic cells and bacterial reverse mutation assay in Salmonella typhimurium TA98 and TA100 with or without metabolic activation system (S9), respectively. In Allium root growth inhibition test, EC50 value was determined approximately 15 g/L and 0.5xEC50, EC50 and 2xEC50 concentrations of Lg-ext were introduced to onion tuber roots and distilled water and methyl methane sulfonate (MMS, 10 ppm) used as a negative and positive control, respectively. The characteristic effect caused by tested preparations was an increase of mitotic index (MI) in 7.5 g/L and 15 g/L (except 24 and 96 h) and simultaneous decrease of MI in 30 g/L and in MMS. While stickiness, bridges, chromosome laggards and disturbed anaphase-telophase were observed in anaphase-telophase cells, c-metaphase, pro-metaphase, polyploidy and binuclear cells were observed in other cells. Lg-ext was not found to be mutagenic on S. typhimurium TA 98 and TA100 with or without S9. The results were also analyzed statistically by using SPSS for Windows, and Duncan's multiple range tests were performed respectively. These results indicate that Lg-ext exhibits genotoxic activity in A. cepa root meristematic cells but not mutagenic activity in Ames test system.

ÖZET: Bu araştırmada, Brassicaceae familyasına ait olan Alyssum virgatum Nyar. bitkisinin toprak üstü kısımlarının distile su ekstrelerinin (Av-eks) Allium cepa L. kök uçları üzerine antisitotoksik etkisi incelenmiştir. Av-eks'in farklı süre ve konsantrasyonlarının MMS (Metilmetansülfonat)'e karşı kök büyümesi, mitotik indeks ve anafaz-telofaz kromozom aberasyonları üzerine etkileri belirlenmiştir. Allium kök büyüme inhibisyonu testinde Av-eks'in 10 farklı konsantrasyonu (100 gL konsantrasyonları hariç diğer Av-eks konsantrasyonları kök uzunluğunu önemli ölçüde azaltmıştır. Kök uzunluğunu yarıya indiren konsantrasyon olarak kabul edilen EC50 değeri yaklaşık olarak 1,6 gL -1 olarak bulunmuştur. Antisitotoksisite testinde, distile su (negatif kontrol), MMS (pozitif kontrol) ve 10 ppm MMS içeren Av-eks'in farklı konsantrasyonları (0,5×EC50, EC50 ve 2×EC50) 24, 48 ve 72 saat süreyle kullanılmıştır. 10 ppm MMS içeren Av-eks karışımlarında, bitki ekstresindeki konsantrasyon artışına bağlı olarak mitotik indekste önemli bir yükselme gözlenmiştir. Kromozom aberasyon çalışmalarında ise anomali yüzdelerinin pozitif kontrol grubuna göre azaldığı tespit edilmiştir. İğ iplikleri bozulmalarının sebep olduğu kalgın kromozom aberasyonu, diğer anomalilerden daha fazla tespit edilmiştir. Sonuçlar Av-eks'in, A. cepa kök meristematik hücrelerinde, MMS varlığında antisitotoksik ve antigenotoksik aktivite gösterdiğini ortaya koymuştur.Anahtar Kelimeler: Allium test, Alyssum virgatum, antisitotoksisite, antigenotoksisite, Brassicaceae ABSTRACT: In this study, anticytotoxic effects of distilled water extract of Alyssum virgatum Nyar. (Brassicaceae family) aerial parts (Av-ext) was determined on Allium cepa L. root meristematic cells. Effects of different concentrations and treatment periods of Av-ext on root growth, mitotic index and anaphase-telophase chromosomal aberrations, against MMS were determined. In Allium root growth inhibition test, EC50 value was determined by using 10 different concentrations of Av-ext (100 gL -1 , 50 gL -1 , 25 gL -1 , 12.5 gL -1 , 6.25 gL -1 , 3.2 gL -1 , 1.6 gL -1 , 0.8 gL -1 , 0.4 gL -1 ve 0.2 gL -1 ). All of the Av-ext concentrations reduced the root length significantly, except 0.8 gL -1 , 0.4 gL -1 and 0.2 gL -1 doses. EC50 value that was accepted as reducing concentration of the root length about %50, was found about 1.6 gL -1 . In anticytotoxicity test, distilled water (negative control), MMS (positive control) and different concentration of Av-ext (0.5×EC50, EC50 ve 2×EC50) including 10 ppm MMS were used for 24, 48 and 72 hours treatment period. In Av-ext mixtures including 10 ppm MMS, it was observed that mitotic index was increased significantly related to the rising concentrations of the plant extract. In chromosome aberration study, percentages of anomallies were decreased according to positive control group. Laggard chromosome aberrations caused by mitotic spindle disruptions were determined more than the other anomalies. The results indicated that Av-ext. has anticytotoxic and antigenotoxic...
