“…With few exceptions, the present findings are similar to those produced by other investigators using salicylates in multiple dose paradigms in rats (Baba et al, 1966; Schardein et al, 1969; Beall and Klein, 1976; Lubawy and Burriss Garrett, 1977; Nakatsuka and Fuji, 1979; Mankes et al, 1982; Hamed et al, 1994). Baba et al (1966) produced two MDs at a 330‐mg/kg dose administered from GD 9 to 14.…”

“…Baba et al (1966) produced two MDs at a 330‐mg/kg dose administered from GD 9 to 14. Lubawy and Burriss Garrett (1977) administered ASA at doses of 125 and 250 mg/kg from GD 8 to 19 to rats. Decreased maternal body weights, increased numbers of resorptions accompanied by decreases in the number of viable fetuses, and decreased fetal and placental weights were noted at 250‐mg/kg dose level.…”

“…40 Lubawy and Garrett treated rats once daily with paracetamol at doses 125 or 250 mg/kg/day between days 8 and 19 of rat pregnancy and did not observe any signs of developmental toxicity. 41 However, when paracetamol was administered three times a day it caused intrauterine growth retardation, reducing fetal weight and length at doses 50, 100 and 200 mg/kg, whereas the placental weight and tail length were increased in the highest drug-treatment group. 42 Data on maternal toxicity, pre-and postnatal losses and bone changes were similar to those in the present study.…”

“…Biologic plausibility stems from animal models, which have demonstrated that reduced fetal length in rat pups is associated with mid-gestation maternal exposure to 125 mg and 350 mg of acetaminophen, dosage comparable to maternal use. (12, 13) Various concentrations of acetaminophen have been associated with morphological changes in the fetal liver. It has been hypothesized that insults to the fetal liver may result in the reduction of blood stem cells to key organs, resulting in increased risk of adverse growth and development.…”
