This study investigated the hypothesis that Portulaca oleracea L. exerts antiobesity and antidiabetic effects by evaluating blood lipid profiles, blood glucose control factors, protein expression of lipid metabolism, and insulin sensitivity improvement. Three groups of high-fat diet (HFD) induced obese C57BL/6 mice (n = 8) received treatment with low (5%; HFD + PO5%) or high (10%; HFD + PO10%) concentrations of P. oleracea powder for 12 weeks or no treatment (HFD) and were compared with each other and a fourth control group. Weight gain was reduced by 34% in the HFD + PO10% group compared to the HFD group. Moreover, the perirenal and epididymal fat contents in the HFD + PO10% group were 6.3-fold and 1.5-fold, respectively, lower than those in the HFD group. The atherogenic index (AI) and cardiac risk factor (CRF) results in the P. oleracea-treated groups were significantly lower than those in the HFD group. The homeostasis model assessment of insulin resistance (HOMA-IR) levels was lower in the HFD + PO10% group than in the HFD group. The protein expression levels of the proliferator-activated receptor (PPAR)-α, glucose transporter (GLUT) 4 and PPAR-γ were upregulated in the HFD + PO10% group compared to the HFD group. However, the protein expression levels of tumor necrosis factor (TNF)-α were lower in the P. oleracea-treated groups than in the HFD group. Our results demonstrate that P. oleracea powder could be effectively used to treat and prevent obesity and diabetes-associated diseases through suppression of weight gain and reduction in body fat and blood glucose levels.

BACKGROUND/OBJECTIVESTurmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet.MATERIALS/METHODSLeptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration.RESULTSContents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group.CONCLUSIONSBased on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.

We investigated the effects of an ethanol extract of Eriobotrya japonica Lindl. (loquat) leaves (EJ) on the lipid metabolism of serum, liver, and adipose tissue, and antioxidative activity in rats fed a fat/cholesterol diet for four weeks. Male Sprague-Dawley rats weighing 207 g were divided into 4 groups: a normal diet group (N), a high-fat/high-cholesterol diet group (HFC), a high-fat/high-cholesterol diet group administered 200 mg/kg day EJ (HFC-EJL), and a high-fat/high-cholesterol diet group administered 400 mg/kg/day EJ (HFC-EJH). The serum ALT and AST activities of the EJ groups were lower than those of HFC group, but there was no significant change in serum ALP or LDH activities. The serum total and LDL-cholesterol, atherogenic index, and cardiac risk factor tended to be decreased in the EJ groups compared to the HFC group, while the serum HDL-cholesterol decreased in the HFC group and increased only minimally in the EJ groups. The total cholesterol in liver and mesenteric adipose tissues was lower in the EJ groups than in the HFC group. Triglycerides in the mesenteric and epididymal adipose tissues were lower in the EJ groups than in the HFC group. The liver GSH levels of the EJ groups were significantly lower than the HFC group. The liver TBARS content was significantly lower in the EJ groups than in the HFC group. These results suggest that EJ ethanol extract may improve the lipid metabolism of serum, liver, and adipose tissue and prevent oxidative stress by stimulating antioxidative systems in rats fed a high-fat/high-cholesterol diet.
