Effect of Staining Procedures on the Results of Micronucleus Assays with Exfoliated Oral Mucosa Cells

Abstract: Micronuclei in exfoliated epithelial cells are widely used as biomarkers of cancer risk in humans. To elucidate the effect of different staining procedures on the outcome of such investigation, we conducted a study in which the micronuclei frequencies in oral mucosa cells of heavy smokers (n = 20) and nonsmokers (n = 10) were evaluated with nonspecific (Giemsa, May-Grü nwald-Giemsa) and DNA-specific (4 ¶,6-diamidino-2-phenylindole, Feulgen, acridine orange) stains, whereas with Giemsa-based stains, the frequen… Show more

“…Cell morphology of Hep-2, HeLa, HT-29, A-549, A-375, HEK-293 and MRC-5 lines was monitored using an inverted microscope under a conventional Giemsa staining protocol (Nersesyan et al 2006). Changes in the cellular morphology were observed and documented after 24 h ExtHS and ExtAS treatment exposure at its IC 50 , The negative control group was treated with EtOH-H2O 70% (v/v), respecting the residual amount ethanol of 3% for ExtHS and destilate water for ExtAS for the same period of time.…”

“…Ayyad SB et al, and Bouter R et al, did not support the use of Feulgen stain for MNi analysis under field condition for the reason i.e., long staining time (>4 hours) and at the same time its method of fixation is more difficult [32,33]. The staining components are expensive and this stain alone can't delineate the cytoplasm; it needs 1% light green to demarcate the cytoplasm and also the hydrolysis is the critical part in Feulgen staining.…”

“…The reliable results were achieved with Feulgen without counterstain (Nersesyan et al, 2006). This technique consists in a first step of hydrolysis with HCL 5M followed by washing with distillate water, incubation with Schiff Reagent and tap water final washing.…”
