Effect of Splenopentin (SP-5) on the Antibody Formation in Immunosuppressed Mice

Abstract: Sublethally irradiated C57Bl/6 mice (whole body irradiation with 600 cGy) were treated with or without diacetyl splenopentin and compared for their capacity to produce antibodies against target sheep red blood cells. As demonstrated, mice treated with splenopentin produced antibodies earlier and in a higher level than animals untreated.

“…In early studies about the influence of splenopentin on the reconstitution of immune reactions after immunosuppression (Diezel et al, 1986;Eckert et al, 1986) as well as on the engraftment after syngenic bone marrow transplantation (Eckert et al, 1989a) a helpful but never a harmful influence was found in every case. Furthermore, the GVHR-induced i1nmunosuppression in the adult H-2 different hybrid model was significantly diminished (Eckert et al, 1989b).…”

“…DAc-SP5 accelerated the reconstitution of antibody formation in vivo after sublethal irradiation (Diezel et al, 1986) and after syngenic bone marrow transplantation (Eckert et al, 1989) as well as in cell transfer experiments in which immunocompetentdepleted spleen cells were transferred into irradiated syngenic mice (Eckert et al, 1986). Furthermore, the symptoms of a graft-vs-host reaction (GVHR) were diminished in adult Fl-hybrids (Eckert et al, 1989).…”

“…Audhya et al (1984) proposed to call this peptide splenopentin (SP-o). The biological activity of SP-o was investigated in vitro in relation to its activity in inducing both Thy-1+ and Lyb-2.1+ antigens on lymphocytes (Audhya et aI., 1984), in affecting peripheral T cells to produce inferferon-gamma (Diezel et al, 1984;Rentz et a]., 1984) and in accelerating the reconstitution of the antibody response (Eckert et al, 1986;Diezel et al, 1986).…”

“…In a number of experiments the immunostimulating effect of splenin was demonstrated in immunosuppressed animals. In all these experiments the animals were treated with splenin 3 times per week up to the day of immunization (Diezel et al, 1986;Eckert et al, 1986;Eckert et al, in press).…”
