Kombucha, also known as the Manchurian mushroom, is a symbiotic culture of bacteria and yeast, the so-called SCOBY. This paper presents a comprehensive evaluation of the ferments obtained from green coffee beans after different fermentation times with kombucha. Results for the ferments were compared to the green coffee extract that was not fermented. In this study, the antioxidant potential of obtained ferments was analyzed by assessing the scavenging of external and intracellular free radicals and the assessment of superoxide dismutase activity. Cytotoxicity of ferments on keratinocyte and fibroblast cell lines was assessed as well as anti-aging properties by determining their ability to inhibit the activity of collagenase and elastase enzymes. In addition, the composition of the obtained ferments and the extract was determined, as well as their influence on skin hydration and transepidermal water loss (TEWL) after application of samples on the skin. It has been shown that the fermentation time has a positive effect on the content of bioactive compounds and antioxidant properties. The highest values were recorded for the tested samples after 28 days of fermentation. After 14 days of the fermentation process, it was observed that the analyzed ferments were characterized by low cytotoxicity to keratinocytes and fibroblasts. On the other hand, the short fermentation time of 7 days had a negative effect on the properties of the analyzed ferments. The obtained results indicate that both green coffee extracts and ferments can be an innovative ingredient of cosmetic products.

Coffea canephora Bean Extract Induces NIH3T3 Cell Migration

BACKGROUND: Wound healing is an essential biological process that consists of sequential steps aimed at restoring the architecture and function of damaged cells and tissues. There are empirical evidences of using pure coffee bean powder as an alternative medicine in treating various types of wounds. However, there is limited data on coffee-induced wound healing, especially migration of cells. Therefore, current study was conducted to investigate the role of coffee extract in cell migration, especially fibroblast which is important for wound healing.METHODS: Coffea canephora beans were prepared, extracted and added in the NIH3T3 cell culture in final concentration of 2.5% and 5%. Then cytotoxicity test was performed using Na,30-[(phenylamino)-carbonyl]-3,4-tetrazolium]-bis(4-methoxy-6-nitro) benzenesulfonic acid hydrate (XTT) assay. Meanwhile, cell migration test was performed with scratch assay. All results were statistically analyzed.RESULTS: The 2.5% or 5% Coffea canephora beans extract (CCBE)-treated NIH3T3 cell numbers were almost similar with the numbers of NIH3T3 cells in starvation medium merely. Meanwhile, 2.5% and 5% CCBE showed significant decrease of the widths of scratched areas compared to starvation medium merely (ANOVA with LSD Post-hoc, p=0.000). After 24 h and 48 h, the average widths of 2.5% and 5% CCBE-treated scratched areas were 235.68±22.79, 50.36±5.29, 229.95±23.01, 27.68±2.83, respectively.CONCLUSION: Since both 2.5% and 5% CCBE are potential in inducing migration of fibroblast (NIH3T3 cell) and do not induce cytotoxicity, the CCBE could be potential as an agent for wound healing.KEYWORDS: coffee, Coffea canephora, NIH3T3, migration, cytotoxicity

Skin graft is a reconstructive procedure commonly used to cover defects or wounds. The majority of post-skin graft treatments use topical agents or the application of modern dressings. However, these treatments have been considered to have disadvantages, such as being limited and expensive. Abundant natural ingredients can be used as alternatives to aid the healing process, one of which is robusta coffee. Robusta coffee is reported to contain active ingredients such as CGA, flavonoids, and saponins. This study aimed to prove the wound healing effect of robusta coffee extract in gel dosage form on the skin graft model in rats. Thirty male Long Evans rats were divided into three groups: negative control (gel base), positive control (1% kojic acid gel), and 5% robusta coffee extract gel (RCEG). A 2 x 2cm split-thickness skin graft was harvested from the dorsal skin of the rat and then fixed to the split-thickness wound at the donor site. Skin graft tissues were taken by biopsy on days 7, 14, and 21 for histopathological evaluation by assessing angiogenesis, fibroblasts, collagen density, and epithelial thickness. Our results found that the number of blood vessels, fibroblast cell count, collagen density percentage, and epithelial thickness were significantly higher in the 5% RCEG group on days 7, 14, and 21 compared to the control group. In conclusion, topical application of 5% RCEG can enhance wound healing of skin graft by improving angiogenesis, fibroblasts, collagen density, and epithelial thickness.
