Effect of Gambisan on the Inhibition of Adipogenesis in 3T3-L1 Adipocytes

Abstract: This study was conducted to explore the antiadipogenic effect and possible mechanism of Gambisan on 3T3-L1 cells. For quality control, Gambisan was standardized by HPLC and the standard compounds ephedrine, epigallocatechin-3-gallate, and caffeine were screened. Cultured 3T3-L1 cells that had been induced to differentiate were treated with various concentrations of Gambisan or its major component extracts (Ephedra intermedia Schrenk, Atractylodes lancea DC., and Thea sinensis L.) for 72 hours for MTT assay to … Show more

“…28 To further our understanding of the effects of OPE on lipid metabolism and adipogenesis, we analyzed the mRNA levels of adipogenic genes and transcription factors that act as master regulators of adipogenesis and lipid storage in 3T3-L1 cells. 29 The differentiation of preadipocytes into adipocytes requires a variety of effectors that activate the process, such as PPAR and C/EBP . PPAR and C/EBP together promote terminal differentiation by activating the transcription of genes crucial for maintaining the adipocyte phenotype.…”

“…Therefore, concentrations less than 0.5 mg/mL were used in all subsequent in vitro experiments. Previous studies regarding antiadipogenic effect of plant extracts in cell lines have also considered the cytotoxicity levels prior to differentiation, focusing on the safety of natural compounds as potential nutraceuticals or functional foods against obesity (Kang, Nam, Kim, Huh, & Lee, 2013;J. Moon, Do, Kim, & Shin, 2013).…”

“…Anti obesity drugs that are efficacious and have minimal side effects are therefore urgent 23 . In this study, we observed the MPEE abilities to prevent the obesity as well as the pathogenesis of metabolic syndrome, by measuring the triglycerides, LDL and HDL cholesterol level.…”
